| Objective : through the tracheaperfusion silica mixed suspension copy silicosis rat model,by observing the Yifei Huaxian prescription effects on the content of HYP and TGF-β1 in the silicosis lung tissue and TGF-β 1 in the silicosis serum,in order to supplementing Qi,nourishing Yin,resolving phlegm and promoting blood in the influence on the occurrence of silicosis fibrosis development and discuss the possible mechanism of action.Methods : 96 healthy male SD rats,were randomly divided into 6 groups,with 16 each group,respectively,normal saline control group(group A)、model group(group B)、low dose Yifei Huaxian prescription group(group C)、middle dose Yifei Huaxian prescription group(group D)、high dose Yifei Huaxian prescription group(group E)and tetrandrine group(group F).Each group on numbers 1 to 16.Group B、C、D、E、F with 0.2g/3ml silica mixed suspension 0.5ml intracheal instillation one time,group A tracheal perfusion amount of saline solution.After 2 weeks,group A、C、D、E began with saline、6g crude drug/kg、12g crude drug/kg、24g crude drug/kg by gavage one time every day,group F with normal tetrandrine 50mg/kg by gavage once every three days,duration 14 days,take number 1 to 8 executed in each group,detection of HYP and TGF-β1 in lung tissue by ELISA,and detection of TGF-β1 in serum.The remaining rats were continuously fed in accordancewith the above methods,duration 4weeks,then serum and lung tissue were measured by the same method.Results:1.model replication:the rats model of silicosis was successfully reproduced by pathology.2.TGF-β1 in serum:the drug was administered for 2weeks and for 6 weeks,low dose Yifei Huaxian prescription group、middle dose Yifei Huaxian prescription group、high dose Yifei Huaxian prescription group and tetrandrine group TGF-β1 in serum were lower than model group,the difference is of significance in statistics science(P<0.05 or P<0.01).3.TGF-β 1 in lung tissue :(1).there was no significant difference between the low dose Yifei Huaxian prescription group and the model group at 2weeks(P>0.05),middle dose Yifei Huaxian prescription group、high dose Yifei Huaxian prescription group and tetrandrine group TGF-β1 in lung tissue were lower than model group,the difference was statistically significant(P<0.01).(2).The drug was administered for 6 weeks:the low dose Yifei Huaxian prescription group 、 middle dose Yifei Huaxian prescription group、high dose Yifei Huaxian prescription group and tetrandrine group TGF-β1 in lung tissue were lower than model group,the difference is of significance in statistics science(P<0.05 or P<0.01).4.HYP in lung tissue:the drug was administered for 2 weeks and for 6 weeks,the low dose Yifei Huaxian prescription group、middle dose Yifei Huaxian prescription group、high dose Yifei Huaxian prescription group and tetrandrine group HYP were lower than model group,the difference is of significance in statistics science(P<0.05 or P<0.01).Concllusion:1.The level of TGF-β1 and HYP in lung tissue andTGF-β1 in serum of silicosis rats were related to the degree of silicosis.2.Yifei Huaxian prescription through the method of supplementiong Qi,nourishing Yin,resolving phlegm and promoting blood circulation can delay the development of silicosis. |
