Effects Of Bufeikangxian Decoction To Expression Of Transforming Growth Factor-β₁(TGF-β₁) MRNA In Rats Of Pulmonary Fibrosis.

Objective: To detect bufeikangxian decoction's effects to expression of transforming growth factor and it's gene in bleomycin-induced pulmonary fibrosis in rat model. To approach the possible mechanism of it's healing pulmonary fibrosis.Methods: 60 male Wistar rats were randomly divided into 4 groups: bleomycin group, control group, bufikangxiang decoction group and prednisone group. Every time 5 rats were put to death after infusing bleomycin 7d, 14d, 28d in each group. Trachea Dynamic changes of the pathomorphilsm changes, the contents of HYP of rats' lung were observed respectively in each group. Bronchoalveolar lavage fluid was collected for ELISA and in-situ hybridization determination.Results: Bufeikangxian decoction has the effects of inhibit the content of TGF-β1 protein, the content of Hydroxyproline(HYP) and abnormal increasing expression of TGF- β 1mRNA and decrease alveolitis and pulmonary fibrosis in degree. TGF- β 1 mRNA expression of bleomycin group was obviously increased compared with the control group. There is a highly significant difference between the 2 groups (P<0.01). After Bufeikangxian decoction or Prednisone administration, an obvious decreasing of TGF- β 1 mRNA was found . Compared with the bleomycin group, there is a highly significant difference (P<0.01, P<0.05). There is not significant difference between the two healing groups.Conclusions: The study shows that Bufeikangxian decoction was effectively improved the symptom of experimental pulmonary fibrosis rats and postponed its proceeding, decreaseing the production of collagen protein,alleviating alveolitis and pulmonaryfibrosis. This may be one of the mechanism of antifibrosis by bufeikangxian decoction .
