The dried rhizomes of Drynariae Rhizoma are known as "Gu sui bu",is one of the traditional Chinese medicines for the treatment of bone disease in China.It can reinforce the kidney,strengthen the bone,promote the healing of fractures,and relieve pain.Rapid analysis and its quality control method were investigated in details by chemical fingerprinting methodology.The thesis is divided into four sections:The first part of this paper summarized the chemical components and the advances of modern pharmacological research.A serious of systematic studies-the processing procedure,variety regulations before and after the processing was introduced.Quality control of Chemical synthesis synthetic material and TCMs by Ambient MS were summarized.In second part,a HPLC fingerprint method was developed for the analysis of Drynariae Rhizoma,and the chemical constituents of Drynariae Rhizoma were confirmed by HPLC-MS.The fragmentation of naringin,neoeriocitrin,caffeic acid 4-O-?-D-glucopyranoside were deduced.Naringin and neoeriocitrin had shown[M-H-120]-ion was principally produced by a reto-Diels-Alder reaction in ring C.To achieve the best effect of extraction,we optimized the condition of the extraction time,extraction of solvent of Drynariae Rhizoma.Combining HPLC fingerprint with Chempattem,the results showed that the difference of chemical composition information between Drynariae Rhizoma and its adulterants.Similarity analysis of Drynariae Rhizoma is greater than 0.75,adulterants less than 0.4.In the principal component analysis,PC1 plus PC2 score of 88.6%,Drynariae Rhizoma and its adulterants were divided into two groups,and were identified accurately.In the third part,a paper spray mass spectrometry method was established for rapid analysis of Drynaria Rhizoma.The authenticity and quality control of Drynariae Rhizoma were studied.In this paper,we optimized conditions for the spray voltage and cone voltage of MS.To achieve the best response and get more chemical information,we compared positive and negative mode used naringin,neoeriocitrin and 4-O-?-D-glucopyranosyl caffeic acid as indicators.The stability of the mass spectrometry was investigated.The intra-day precision was not more than 5.28%and the intra-day RSD was less than 7.27%.In the negative ion mode,the m/z 191,341,377,579,595,615,631 in the traditional Chinese medicine have the best response,and the m/z 191,341,371,533,609 are better in adulterants.The peaks of 4-O-?-D-glucopyranosyl caffeic acid were also shown in adulterants,and the peaks of the naringin and neoeriocitrin were not detected in adulterants.In principal component analysis,m/z 579,595,615,631 contribution to the largest,30 samples will be divided into two groups.The method of paper spray mass spectrometry greatly shortens the time of analysis,achieving the purpose of e identification rapidly and accurately.It is expected to be used for rapid identification of large quantities of medicinal materials.The fourth part compared the changes of the chemical information of microwave processed products and the hot in sand products and raw materials by HPLC,PS-MS,HPLC-MS.The results showed that compared to raw materials,Drynariae Rhizoma produce 5-hydroxymethyl furfural after microwave processing and sand processing,a kind of stimulating ingredients.The proposed method need a reasonable processing temperature.Dissolution rate of three main active ingredients,4-O-?-D-glucopyranosyl caffeic acid,naringin and Neoeriocitrin were increased after sand processing and microwave processing.Dissolution rate of three main active ingredients were 12.54%10.97%and 8.16%respectively after hoted in sands.Paper spray mass spectrometry combined with Chempattern analysis,raw medcine and processed medicine were divided into two groups,indicating that the chemical information of Drynariae Rhizoma has changed after the processing technology which played an important role in quality control of processed medicine.
