Objective: This experiment research by observing qilongtongshentang in the treatment of membranous nephropathy model rats urinary protein,kidney pathological structure,quantitative glomerulus in membrane protein expression difference of Nephrin and Podocin,explore qilongtongshentang treatment of kidney disease model rats curative effect and action mechanism of proteinuria.Methods: Choose clean level 72 healthy male SD rats and adaptability to feed a week,then randomly selected twelve as blank control group,the rest of the rat,s caudal vein and test low,medium and high dose groups according to random indicator method,each group of twevel.Soup of stilbene dragon pass kidney oral dose of 126 g * 1/60 adults 1 kg = 2.1 g/kg,low,medium and high dose group of rats were given Qilong Tongshen Tang of 15.75g/kg/d(adult doses of 7.5 times),31.5 g/kg/d(adult dose of 15 times),63 g/kg/d(30 times the dose of adult)to fill the stomach;Leigongteng Duodaipian as a positive control group,adult day oral dose of 1 mg/kg/d,give the dose of rats by 30 times the dose of adult or 30 mg/kg/d,prepared with distilled water as the suspension liquid lavage,blank control group and model control group such as volume distilled water to fill the stomach,once per day,for four consecutive weeks.Each group had a stomach volume of 20ml/kg/d.After the treatment,then observe the changes of rat 24 hours urinary protein quantitative;Abdominal aortic blood,centrifugal supernatant,detect each group rats serum total protein(TP),albumin(propagated),total cholesterol(T-CHOL),triglyceride(TG),serum creatinine(Scr),the change of urea nitrogen(BUN);Application of the double antibody sandwich ELISA method for determination of TGF-beta 1,the change of IL-6.In rat kidney,kidney changes under light microscope and electron microscope observation,using immunohistochemical method to detect the rat glomerular podocyte changes in membrane protein Nephrin and Podocin expression.Results:1.qilongtongshentang can decrease the membranous nephropathy(MN)in the rat quantitative 24 hours urinary protein,improve the level of TP,propagated,reduce T-CHOL,TG level.2.qilongtongshentang can decrease the MN rats serum TGF-beta 1 and the content of IL-6,compared with model group difference was statistically significant.3.qilongtongshentang can improve MN Nephrin and Podocin rats,to maintain the integrity of the cell shape function,reduce proteinuria leakage.4.qilongtongshentang capsule can reduce the immune complex deposition in GBM,lighten the GBM thickening,thus reduce the MN rat renal pathological damage.Conclusion:Qilongtongshentang can effectively reduce the MN nephropathy rats proteinuria,alleviate MN rat renal pathological damage,by raising MN rat glomerular podocytes in the expression of membrane protein Nephrin and Podocin.
