1 research purposesThe purpose of this experiment was to compare from animal behavior and sex hormones E2,T and PRL in two aspects of Xiaoyao Powder Free Decoction and Xiaoyao hollow suppository provide support.2 research methodologyIn January at the age of 50 female SD rats were randomly divided into 5 groups: control group,depression model group,Xiaoyao Decoction group,Xiaoyao granules hollow suppository high dose group,low dose Xiaoyao hollow suppository group,10 rats in each group,through a variety of chronic unpredictable stress depression stimulation random combination of Ganyu sex hormone testThe blood from abdominal aorta was centrifuged and the serum was separated.The contents of E2,T and PRL in serum were determined by radioimmunoassay.3.5 statisticsData were analyzed using SPSS17.0 software,the measurement data to mean +standard deviation,comparison between groups using independent sample t test,were compared with paired t test,analysis of variance was used to compare quantitative data between groups,P < 0.05,the difference was statistically significant.4 Results4.1 the successful model of decision: compared with the blank control group,model group,mean free Decoction group,Xiaoyao Xiaoyao hollow suppository high dose group,low dose group of Xiaoyao hollow suppository of general status,body weight,feeding behavior and the observation of a significant change in liver after modeling,analysis by t test,there was statistical the significance of P is less than 0.05 difference,suggesting that the model was successful.4.2 Observation ofgroups of vaginal smears: model control group after 5 days of smear in proestrus,estrus,estrous period of rest interval,alternately,only 1 days under the microscope showed typical keratinized cells increased;Ganyu > 0.05,no difference between the two groups.4.4 based on the sex hormone E2,T,PRL detection results: Xiaoyao Powder Free Decoction group,Xiaoyao hollow suppository high dose group T,PRL and liver of model group compared to P < 0.05,there was statistical significance;Xiaoyao hollow suppository low dose group PRL with Ganyu model group compared to P < 0.05,there was statistical significance;the comparison between groups by statistical analysis P > 0.05,that there is no difference.5 Conclusion5.1 the rats of various stress 21 days chronic unpredictable depression stimulation random combination method,can successfully replicate the Ganyu model.5.2 Ganyu model female rats with poor general condition,weight loss,decreased food intake,estrous cycle disorder,open field test in the horizontal and vertical score decreased the elevated plus maze test in the open arm time and frequency ratio decreased,sucrose preference decreased.5.3 happy hollow suppository can improve the liver qi stagnation syndrome in female rats general state,weight gain,food intake increased,raised in the open field test of the horizontal and vertical score,elevated plus maze open arm time and frequency ratio,sucrose preference degree and improve the disorders of estrous cycle,serum E2,T regulation and PRL content.Free decoction had no significant differences compared with Xiaoyao Powder,can be used as a new dosage form for clinical application.
