Objective: To explore the profile expression of long non-coding RNA(lncRNA)of stable angina pectoris(SAP)by using gene chip and bioinformatics technique.Screen the specific lncRNAs of stable angina pectoris,explore preliminary information of biological functions and discovere new molecular markers for the diagnosis of coronary heart disease from the differences in the expression of lncRNAs.Methods: 5 patients with stable angina pectoris and 5 healthy people(non-SAP)were selected,then drew the blood.Blood samples were extracted to RNA.The expression of lncRNA and microRNA(mRNA)were analyzed by gene microarray.The differential expression profiles of lncRNA and mRNA were established by means of scatter plot,volcanic map and cluster analysis.Screened the specific high expression of lncRNAs according to FC(abc)value> 2,P <0.05.Conjointly analysis the differentially expressed lncRNAs by using bioinformatics methods.Including: KEGG Orthology Based Annotation System(KOBAS)analysis and Gene Ontology(GO)analysis;lncRNA-mRNA association analysis.So as to predict the biological function may be involved in the differential expression of 1ncRNAs.Results: The expression of 1ncRNA and mRNA in SAP were compared with non-SAP.The quantity of differently expressed lncRNAs and mRNA were 22154 and 25138 respectively.SAP group was 399 lncRNAs according to FC(abc)value> 2 and P <0.05,and 226 lncRNAs were significantly up-regulated,while 173 lncRNAs were down-regulated.According the same standard,a total of 458 mRNAs were screened,which 347 were up-regulated and 111 were down-regulated.GO function enrichment included molecular metabolism(energy,zinc ion metabolism,etc.),cathepsin,tumor necrosis factor reactivity,oxidative stress conduction,negative regulation of histone and other related functions.Signal pathway enrichment contained EGF receptor regulation,DAG and IP3 regulation,HSF1 activation,physiological rhythm,protein carboxylation,transport,division,Akt activation PI3 K and other related pathways.Enrichment of the disease included metabolic diseases,hypertension,arterial disease,calcium block related diseases,cognitive dysfunction and other related diseases.Conclusions: In stable angina pectoris,lncRNA and mRNA are significantly up-regulated or down-regulated,suggesting that these differentially expressed lncRNAs were may involved in some regulatory processes for the development of stable angina pectoris.Highly expressed lncRNAs laid the foundation for development as clinical diagnostic markers.Through further biological function information exploration,the expression of differentially expressed lncRNAs were involved calcium channel,molecular metabolism,myocardial contractility,hypertension and other biological functions and a variety of cardiovascular diseases,reflected the different expression of lncRNAs were closely related to SAP.Provided the theoretical basis for the follow-up study of coronary heart disease.
