Objective: The potential effects of polysaccharide,Shubat and the combination on PM2.5-induced lung injury in rats were explored.Method: 104 Wistar rats were randomly divided into control group,PM2.5 group,Vitis Viniferal L polysaccharides(VTP)high,medium and low dose groups,Lycium barbarum polysaccharide group(LBP),Shubat high,medium and low dose groups,fermented cow milk,combined high,medium and low dose groups.The control group treated nothing,PM2.5 group treated with distilled water,the other groups give the corresponding substances for 30 days,rats in each groups instilled with PM2.5 suspension into trachea except the control group.Blood and bronchoalveolar lavage fluid(BALF)were collected after 24 h.Alkaline phosphatase(ALP),lactate dehydrogenase(LDH),tumor necrosis factor-?(TNF-?),interleukin-6(IL-6)in BALF,ALP and LDH in surem,superoxide dismutase(SOD),malondialdehyde(MDA)in homogenate were detected.Calculating the ratio of wet and dry weight(W/D)of lung tissue,and spleen,thymus,lung indexes.Taken the lung tissue for pathological examination.Result: Compared with the PM2.5 group,(1)there was no difference between each groups in body weight.(2)The spleen index in VTP groups and combined high-dose group were elevated(P<0.05),there was no difference between each groups in thymus index(P>0.05).The lung index in VTP middle and high-dose groups,Shubat middle and high-dose groups,combined middle and high-dose groups were declined(P<0.05).(3)The ratio of wet and dry weight of lung was decreased except VTP low-dose group(P<0.05).(4)ALP and LDH in serum and BALF were decreased in VTP,Shubat and combined groups(P<0.05).(5)In VTP,Shubat and combined groups,SOD increasedwhile MDA decreased(P<0.05).(6)TNF-? in VTP and Shubat groups decreased(P<0.05),there was no significant difference between the combined groups and the PM2.5 group(P>0.05).IL-6 was elevated in high-dose VTP group and decreased in high-dose Shubat group(P<0.05),there was no significant difference between the combined groups and the PM2.5 group(P>0.05).(7)Pathological changes: Alveoli and bronchial wall were intact in control group.In PM2.5 group,the alveolar septum was widened and there was inflammatory cell infiltration in the bronchial wall and pulmonary interstitium.In VTP,Shubat and combined groups,alveolar septal thickening decreased in varying degrees,inflammatory cells decreased in varying degrees.Conclusion: VTP,Shubat and the combination could protect the rats lung injury induced by PM2.5,mainly appears in the improvement of lung permeability,oxidative stress,inflammation,tissue edema and pathology.
