Objective:To explore the impact of polysaccharides from vitis vinifera L.(VTP)on learning and memory ability in Alzheimer's disease(AD)rats and the potential protective effects.To explore the effect of VTP on hippocampal neurons of Alzheimer's Disease(AD)model rats.Methods:Ninety male Sprague-Dawley rats were randomly devided into control,model,positive control group(donepezil group,0.5mg/kg),VTP low-dose,middles-dose and high-dose(300mg/kg,150mg/kg,50mg/kg).A?25-35 was injected into the bilateral hippocampal CA1 to induce AD models.The Morris water maze and Y-maze was used to investigate the ability of learning and memory.The supemxide dismutase(SOD),glutathione peroxidase(GSH-Px),catalase(CAT)activity and malonaldehyde(MDA)content in serum and hippocampus were measured by reagent kits.The Neurons morphological changes of hippocampal CA1 neurons was observed by light microscope and electron microscope.TUNEL staining was used to detect the apoptosis of hippocampal neurons.Results: Compared with those of model group,the learning and memory abilities of VTP and Donepezil groups were significantly improved(P < 0.05),the number of nerve norml cells was significantly increased and apoptosis cells was significantly decreased(P < 0.05).In comparison with those of control group,Y-maze error number and standard time was significantly increased(P<0.05),serum and hippocampus GSH-Px,SOD,CAT activity was significantly decreased(P<0.05),MDA content was significantly increased(P<0.05)in model group.Compared with those of model group,error number and standard time of Y-maze was significantly reduced(P<0.05),serum and hippocampus GSH-Px,SOD,CAT activity was significantly increased(P<0.05),MDA content was significantly reduced(P<0.05)in high and medium VTP dose groups.Rats in the control group had no obvious morphological changes,and morphology of rat in the VTP and Donepezil groups was basical normal,however,the obvious deposition of cellular morphology was displayed in model group.Conclusion:VTP can improve the ability of learning and memory and protect the hippocampal neurons of AD model rat,through alleviating hippocampal CA1 neurons damage induce by amyloid beta.VTP has the effect of improving the learning and memory ability of AD rats,which could alleviate neurotoxicity of A? via inhibition of oxidation.
