Objective:This study used the method by stomach perfusion of the Diphenoxylate suspension to copy the animal model of constipation,to observe the effect of Zengye Decoction for increasing fluid on defecation in experimental animals,effect on intestinal absorption of water,effects on aquaporin3(AQP3)and aquaporin9(AQP9)of colonic tissue,effects on motilin(MTL)and somatostatin(SS)of the serum and effects on isolated intestine in Rats.On the molecular level,the level of gastrointestinal hormones and the level of receptor,To explore the mechanism of action of Zengye Decoction in the treatment of constipation,to provide scientific basis for Zengye Decoction for treating constipation.Methods:1?60 male and female SPF Kunming mice were divided into the normal group,the model group,the Maren capsules group,the high,the medium,and the low does Zengye Decoction groups 10 mice for each.Established constipation mouse model by gastrogavage Compound Diphenoxylate tablets.After the establishment of the model,with Zengye Decoction intervention,to study the effect of Zengye Decoction on defecation,intestinal water absorption for constipation model in mice.2?60 male and female SPF SD rats were divided into the normal group,the model group,the Maren capsules group,the high,the medium,and the low does Zengye Decoction groups 10 mice for each.Established constipation rats model by gastrogavage Compound Diphenoxylate tablets.After the establishment of the model,with Zengye Decoction intervention,the changes of AQP3,AQP9 and serum MTL and SS in colon tissue were detected by ELISA and the changes of AQP3 mRNA,AQP9mRNA by RT-PCR.3?8 male and female SPF SD rats,fasting water 16 hours,to take off the cervical vertebrae to execute the rats,quickly cut open abdominal cavity,cut a colon,divided into small pieces of sections as a specimen.One end of specimen is fixed on ventilation hook,the other end is connected to the tension converter,connection with Powerlab biological signal acquisition system,to observe the effects of Zengye Decoction and different receptor blockers on the specimens.Results:1?Zengye Decoction can significantly shorten the defecation time and defecation of the model mice,and the difference was statistically significant(P<0.05,P<0.01).2?Zengye Decoction can significantly increase the water content of intestinal tract in mice with constipation,and the difference was statistically significant(P<0.05,P<0.01).3?Zengye Decoction can significantly reduce the content of AQP3 and AQP9 protein in rat colon,and the difference was statistically significant(P<0.05,P<0.01).4 ? Zengye Decoction could decrease the expression of AQP3 mRNA and AQP9 mRNA in colon of rats,but there was no significant difference(P>0.05).5?Zengye Decoction can significantly increase the serum MTL levels,reduce the SS content,and the difference was statistically significant(P<0.05,P<0.01).6?Zengye Decoction can obviously increase the muscle tension of isolated rat intestine,the difference was statistically significant(P<0.05).And have a increase trend in vitro intestinal muscle on join the ? blockers,but the difference was not significant(P> 0.05).Conclusions:1?Zengye Decoction through decrease colon AQP3 and AQP9 content,reduce on intestinal water absorption,increase the intestinal water content,moisten the intestinal tract,at the same time convenient stool discharge,to treat constipation objective.2?Zengye Decoction by increasing the serum MTL content,reduce the serum SS content,to promote gastrointestinal peristalsis and increase intestinal muscle tension,in order to promote rapid feces through the intestinal tract,shorten defecation time,so as to achieve the purpose of treating constipation.3?The mechanism of Zengye Decoction for treating constipation may be achieved by regulatingAQP3,AQP9,MTL,SS and increasing intestinal muscle tension.4?The effect of Zengye Decoction for increasing fluid and moistening liquid to treat constipation is exact,and the clinical effect was the most obvious.
