| Objective:To study the effect of raspberry stem on drug-induced liver injury mice and the expression of HMGB1,TLR4 and NF-κB protein.Provide basic experimental evidence for the further study and the clinical application on liver injury of raspberry stem.Methods:72 healthy male ICR mice,randomly divided into 6 groups:Blank group,Model group,Biphenyl dimethyldicarboxylate(DDB)group,Raspberries stem low dose group,Raspberries stem middle dose group,Raspberries stem high dose group.Model group was given equal volume of normal saline as other groups.Biphenyl double lipid(DDB)group was given biphenyl dimethyldicarboxylate 0.15g/kg.Low,middle and high dose group of raspberry stem was given raspberries stem decoction of 1.14 g/kg,2.28 g/kg,4.56 g/kg by intragastricing administration once per day,four weeks.After the intragastric administration of raspberries stem,we established hepatic injury model by intraperitoneal injecting acetaminophen(APAP)500mg/kg.Observed the general condition of mice before killing them.We got the blood by removalling eyeball for testing the content of ALT,AST.The same section of liver was obtained for HE staining.We got another part of liver for TNF-a and IL-1β detecting.The expression of HMGB1,TLR4 and NF-kappa B was detected by the immunohistochemical determination.Results:HE staining showed obvious liver injury by intraperitoneal injecting APAP in model group.Compared with the blank group,the serum levels of ALT and AST in the model group were significantly increased(P<0.01);Each dose group of raspberry stem could decrease the level of serum ALT,AST at varying degree.Middel and high dose group of raspberry stem could significantly decrease the level of TNF-a and IL-1β(P<0.01,P<0.05).The expression of HMGB1,TLR4 and NF-κB protein could be significantly inhibited by high and middle dose group of rsapberry stem(P<0.01,P<0.05).Conclusion:Raspberry stem could reduce the liver injury by inhibiting the expression of HMGB1,TLR4,NF-kB protein and regulating the level of inflammatory cytokines in liver tissue. |
