Bortezomib is first proteasome inhibitors used in the clinical(The us food and drug administration(FDA)approved it for clinical in 2003),and the inhibitory effect for varied tumor cells has been proved by Many experimental studies.But there is less research on the therapeutic effect of Bortezomib on Colon carcinoma.This paper investigate morphology and particle size of the drug micelle.The results show that the micellar shape is spherical and the particle size is 117 nm.Determined by MTT method in vitro to investigate cytotoxicity of C26 cell line,results show that the nanometer material has negligible cytotoxicity,Bortezomib-loading nanoparticles micelles shows the better cytotoxicity.Application of flow cytometry and laser confocal microscopy testing uptake of C26 cells for drug micelle in vitro,the experimental results show that the uptake of nano micelle is increasing by time.Animal experiments,hematoxylin eosin staining of subcutaneous transplantation tumor biopsies and immunofluorescence staining of Bortezomib-loading nanoparticles micelles shows a better comprehensive cancer treatment effect,and the its mechanism to inhibit tumor growth,promote apoptosis of tumor is through of promoting CHOP and Caspase-3 expression and inhibiting NFk B signal.Above all,loading nanoparticles micelles is a kind of relatively safe and effective new type nano drug on Colon carcinoma.
