Alcoholic liver disease is frequently induced by a long term consumption of alcohol.Alcoholic liver disease was classified according to the characteristics of fat steatosis,hepatitis,liver fibrosis and cirrhosis.Severe hepatitis and cirrhosis are the leading cause of death,and the incidence and mortality of alcohol-induced liver disease are increasing year by year.Now,the prevention and treatment of alcoholic liver disease is either to quit wine or to remedy with drugs.Corticosteroids and pentoxifylline are new commonly used clinical drugs.Most of these drugs,however have no obvious therapeutic effect in alcoholic liver disease examined by pathology of the scoring system.Therefore,developing a novel effective medicine for alcoholic liver injury is of great significance.Ginseng,a natural medicinal plant,has utility in tumor prevention and treatment,eliminating inflammation,relieving aging,enhancing immune activity.Ginsenoside,as main active ingredient of ginseng,has been widely used as a supplemental drug.A number of study showed that ginsenosides have a utility in protecting liver injury.ANTICan is an extract from a specific Jilin Ginseng,which are processed though the reagents and methods approved by the State Food and Drug Administration.ANTICan is rich in ginsenosides Rh4,Rg3,Rg5,Rk1 and Rk3,which are of high activity and high bioavailability.These favorable ginsenosides,surprisingly,accounted for 80% of the total content of ginsenosides,which was 500 times more than that in white ginseng and 12-15 times more than that in the wild ginseng.This thesis focused on the protective effect of ANTICan on alcohol-induced liver injury.To accomplish this objective,we have made the following studies:1)Using high performance liquid chromatography(HPLC)determined the ginsenosides of ANTICan.Among them,the amount of highly active rare ginsenosides Rg3,Rg5,Rh4,Rk1,PPT and PPD was 80% of total ginsenosides.2)The antioxidant ability of ANTICan was determined with DPPH,and 0.25mg/m L of ANTICan removed 50% of free radicals3)MTT assay was used to determine the protective effect of ANTICan on the survival rate of normal liver cells treated with alcohol.6mg/m L ANTICan could effectively protected cells from alcohol injury.4)The model of alcoholic subacute liver injury was established by C57 BL/6 mice.5)ANTICan dose-dependently reduced alcohol-induced increase in aspartate aminotransferase,alanine aminotransferase,alkaline phosphatase,total bilirubin,low density lipoprotein,and total cholesterol.6)Antioxidant enzyme levels in the liver tissue homogenate were tested,and ANTICan dose-dependently increased the alcohol-induced reduction in superoxide dismutase and reduced glutathione.7)Using the kit to detect lipid peroxidation products in the liver tissue homogenate,ANTICan dose-dependently reduced alcohol-induced upregulation in malondialdehyde.8)Pathological analysis of mice liver was performed by HE staining and found that ANTICan reduced the fatty degeneration and the accumulation of inflammatory cells caused by alcohol.9)Fat analysis of mice liver was ferformed by oil red O straining and found that ANTICan significantly reduced the accumulation of triglyceride.Taken together,the present study showed that ANTICan is rich of rare ginsenosides which are of good absorption and high activity,and has a strong antioxidant activity.In vitro,ANTICan can effectively protected human normal liver cells L-O2 damage induced by alcohol.In vivo animal models,ANTICan can also effectively protected alcoholic liver injury.Therefore,we suggest that ANTICan has the potential activity in treating alcoholic liver injury.
