Objective:To observe the alcohol extract of ginseng and its effects on lowering blood glucose in diabetic rats the liver glycogen synthase kinase-3β transcript levels in the initial clarify its mechanism.Methods:In this study, streptozotocin (STZ) induced diabetic rat model, total normal control group, diabetic control group, ginseng ethanol extract of high-dose group, a small dose of ethanol extract of ginseng group of four groups, each10, the experimental period of4weeks. Observed changes in blood sugar, while the use of immunohistochemical staining of pancreatic islet changes in rats, histochemical PAS staining of glycogen content in rats, RT-PCR experimental technique to observe the rats liver glycogen synthase kinase-3β mRNA expression.Results:(1) The blood glucose values, diabetes group, large doses of ethanol extract of ginseng and ginseng alcohol extract group of low-dose serum FBG levels compared to normal control group, there was significant difference (P<0.01); after administration FBG levels of ethanol extract of ginseng, comparing high-dose group compared with the previous administration there was significant difference (P<0.01), ginseng ethanol extract of low-dose group compared with the previous administration was no difference (P>0.05).(2) large doses of ethanol extract of ginseng group than the diabetic control group islet change has changed. Islet irregular shape, state of confusion, the number of immunoreactive cells were markedly reduced, but some cytoplasmic granules of endocrine and diabetes compared with the control group a larger number, color depth, clear lines, the distribution is uniform. Ginseng ethanol extract of low-dose group compared with the diabetic control group was similar to staining.(3) ginseng ethanol extract of high-dose group liver PAS reaction between normal control and diabetic control group, hepatic steatosis was significantly reduced compared with diabetic control group, glycogen content was significantly increased compared with diabetic control group, staining deepened, but the distribution of glycogen granules compared with the normal control group uneven, blurred. Ginseng ethanol extract of low-dose group positive for PAS reaction product of a small liver, slightly more than the diabetic control group.(3), glycogen synthase kinase-3β mRNA expression in diabetic control group and normal control group with significant difference (P<0.01); ginseng ethanol extract of high-dose group and the diabetic control group with significant difference (P<0.01); ginseng ethanol extract of high-dose group and the ethanol extract of ginseng compared with low-dose group were significantly different (P<0.01); ginseng ethanol extract of low-dose and diabetic control group was not statistically significant (P>0.05); normal control group with large doses of ethanol extract of ginseng was no statistically significant (P>0.05).Conclusions:(1) ginseng alcohol extract of STZ-induced diabetic rats has a significant hypoglycemic effect.(2) of ethanol extract of ginseng can promote the regeneration of pancreatic islets in diabetic rats and restoration.(3) of ethanol extract of ginseng can increase the liver glycogen synthesis, this process may be through inhibition of hepatic glycogen synthase kinase-3β to achieve.
