BackgroundsBreast cancer is a malignant tumour of the topmost threat to female health.Although China is not the high incidence area of breast cancer around the world,the incidence of breast cancer in China presents a higher growth rate since the 21 st century.According to China's 2012 reports of cancer registry data(2009),the average prevalence of breast cancer is 42.55 over 100000 in women in China with the first place in female malignant tumours,and the mortality rate is 10.24 over 100000 in women,ranked fifth.Due to the pollution of the environment,lifestyle changing,and many other factors,the incidence of breast cancer in China continues to rise.The latest datas show that the prevalence rate of major cities,including Beijing and Shanghai has exceeded 70 over 100000 in women.Hence,there is a way to go in prevention and cure of breast cancer.Early discovery,early diagnosis and early treatment have important clinical and social significance.In order to achieve this goal,except taking positive and effective prevention and control strategy,we must also make accurate judgment and projections for the biological behavior of breast cancer.In recent years,the development of molecular biology technology deepens our understanding of biological behavior of breast cancer at the molecular level,helping us to find some key molecules such as human epidermal growth factor receptor 2(HER-2),estrogen receptor(ER),progesterone receptor(PR),Ki-67,which play important roles in breast cancer diagnosis,treatment options,prognosis judgement and condition monitoring.PFTK1 is one of the cell cyclin-dependent kinase family.In recent years,with the deepening research on PFTK1,we have discovered PFTK1 is a very potential tumor associated protein,which is closely linked to the occurrence and development of tumor proliferation and metastasis and has the potential to become the new targets for cancer treatment.And vascular endothelial growth factor(VEGF)is the most common form of angiogenic factors.The study find that increasing positive expression rate of VEGF in breast cancer tissue and the expression level and microvascular generation levels are positively correlated.In this study,immunohistochemical Streptavidin-Perosidase(SP)method is used to detect the expression of PFTK1,VEGF and microvascular density(MVD)in breast cancer tissue so as to discuss the relationship between the expression of PFTK1 in breast cancer tissue and microvascular generation.ObjectiveTo explore the expression of PFTK1 in human breast cancer,its relationship with clinicopathological features of breast cancer and its influence on vascular endothelial growth factor(VEGF),microvascular density(MVD).MethodsUsing immunohistochemical SP method to detect the expression of PFTK1 in 113 cases of breast cancer tissue and the tissue adjacent to carcinoma and the expression of VEGF and MVD in breast cancer tissue for statistical analysis.Results1.The expression of PFTK1 in breast cancer tissues is intensity,the total positive rate was 55.8%(63/113),higher than the positive rate in the tissue adjacent to carcinoma,which was 33.6%(38/113).They have statistical significance(P = 0.001).2.VEGF positive expression rate in PFTK1 positive group was 71.4%(45/63)was significantly higher than that in PFTK1 negative groups,which was 26.0%(13/50),the difference is significant(P = 0.000).3.The MVD value of the two groups were 53.04±8.05 and 44.95±8.72,have significant difference(P = 0.000).4.PFTK1 positive expression rate in the groups according to the situation of axillary lymph node metastasis,TNM staging and the expression of Ki-67 have significance statistical differences(P < 0.05),but that in the groups according to age,tumor size,and the expression of ER,PR,Her-2 have no significance statistical differences(P > 0.05).ConclusionPFTK1 may play an important role in micro-angiogenesis in tissue of breast cancer,and may be associated with tumor cell proliferation and metastasis.
