Ovarian cancer is one of the most common malignant tumors in gynecology,Malignant tumor has become a disease in today's punished severely threatening human health.Breast and ovarian cancer is growing rapidly.Ovarian tumors phase I and phase II prognosis is good,but the disease has been found to stage III and VI,the 5-year survival rate of the patient is still hovering at around 35%.Its high fatality rate is the primary cause of early distant metastasis occurred.Has long been surgery combined paclitaxel and platinum based chemotherapy,and intraoperative and postoperative peritoneal perfusion chemotherapy,and targeted new drug application together improve survival,but the effect is not optimistic.Ovarian malignant tumors including epithelial tumors(EOC),germ cell tumor,sex cord stromal tumor,etc.Although many studies for ovarian cancer,but there is not very clear about the pathogenesis of ovarian cancer,there is still no specificity and sensitivity of the screening method.So clearly different classification of ovarian cancer occurrence and development of the molecular pathways,and the new biomarkers to improve the diagnosis and monitoring of ovarian cancer patients to explore the new method is especially important.Fork box protein Q1(forkhead box Q1,FOXQ1)is a member of the fork frame transcription factors family,as a kind of nuclear transcription regulatory factors,involved in embryonic development,biological aging,sugar and lipid metabolism,cell cycle regulation and other biological processes;Its mutation and abnormal expression is associated with tumorigenesis are.FOXQ1 epithelium and stroma is closely related to the transformation process,participate in multiple transformation of epithelial mesenchymal(TGF-beta/Smad signaling pathways,Ras/PI3 k signaling pathways,etc.)related signaling pathway conduction,is a key factor in the process of EMT happened.FOXQ1 the TGF-beta 1 regulating effect on downstream target genes influence the expression of E cadherin and N-cadherin,make interstitial changes of the epithelial cell migration and invasion ability.Transformation of epithelium and stroma(EMT)is present in the phenotype of epithelioid cells lose their polarity,showed enhanced activity ability,able to move freely between the cell matrix and fiber sample phenotype transformation process.The tumor cells can form the local invasion and distant metastasis,is an important factor in patients with postoperative recurrence.This study applies fluorescence quantitative reverse transcription polymerase chain reaction(qRT-PCR)method and immunohistochemical staining method to detect the normal ovarian tissue,benign ovarian epithelial tissue and epithelial ovarian cancer tissues FOXQ1,E-cadherin,N-cadherin,TGF-beta 1 and VEGF mRNA and protein expression of statistical trend,its voice analysis and epithelial ovarian cancer,the relationship between the clinical pathological features of expression and analysis in the above factor correlation each other.ObjectiveThis study aims to observe the fork frame protein Q1(FoxQ1),E-calcium mucins(E-cad)and N-calcium mucins(N-cad),transforming growth factor beta 1(TGF beta 1)and vascular endothelial growth factor(VEGF)in normal,benign and malignant ovarian epithelial tissue of expression,in protein expression level of correlation analysis,and discuss the changes in the existing contributes to an understanding of the significance of tumor occurrence and development process.Materials and methods1.Study objects: Research from from January 2013 to December 2013,third affiliated hospital of zhengzhou university line of surgical treatment of 117 cases of patients with ovarian epithelial tissue,117 patients aged 17 ~ 76 years old,average 48.3 years old.Among them,the epithelium of 40 patients with ovarian cancer(malignant group),39 patients with benign epithelial ovarian tumor(benign group)from patients with benign ovarian tumor appendix resection;Research into the standard: neither preoperative chemoradiation and other auxiliary treatment.All cases were confirmed by surgery and pathology.Ethics committee approval and consent from the subjects are collected.2.Methods: Cut out a small amount of ovarian epithelial tissue,application of fluorescence quantitative reverse transcription polymerase chain reaction(qRT-PCR)method and immunohistochemical staining in the groups of ovarian epithelial tissue was detected in FOXQ1,E-cad,N-cad,TGF-beta 1,VEGF mRNA and protein expression,can use Spearman correlation analysis,the analysis of the correlation between the above indicators.3.Statistics treatment:Adopting SPSS21.0 software for data processing.Normal distribution measurement data using mean ± standard deviation inspection,said the non-normal distribution of measurement data with median(interquartile spacing).Comparison between groups using single factor analysis of variance.Count data comparison with chi-square test;When comparing two test level between groups of??=?/3=0.0167.Inspection level for alpha = 0.05.The correlation between the two variables inspection Spearman correlation analysis.Results1.3 kinds of organization FOXQ1,N-cad,E-cad,TGF-beta 1 and the expression of VEGF mRNA FOXQ1,N-cad,TGF-beta 1,VEGFmRNA in the normal group,the relative expression quantity in benign and malignant group respectively,and 0.901(0.654,0.654),3.099(0.389,0.389)and 0.389(0.310,0.310);1.097(0.712,0.712),3.003(0.334,0.334)and 0.334(0.987,0.987);0.130(0.981,0.981),0.622(0.530,0.530)and 0.530(0.660,0.660);1.310(0.908,0.908),2.009(1.009,1.009)and 1.009(2.098,2.098).Compare the differences between groups were statistically significant(P < 0.05).E-cadmRNA in the normal group,the relative expression of benign and malignant group were 7.912(1.091,1.091),3.880(0.590,0.590)and 0.590(1.250,1.250).Compare the differences between groups were statistically significant(P < 0.05).2.FOXQ1 in three kinds of organization,N-cad,E-cad,TGF-beta 1 and the expression of VEGF protein comparative FOXQ1,N-cad,TGF-beta 1 and VEGF protein expression in normal group of low and middle,high expression in malignant group,compare the differences between groups were statistically significant(P < 0.0167);E-cad high expression in the normal group,lower the expression in malignant group,compare the differences between groups were statistically significant(P < 0.0167).3.In epithelial ovarian sex FOXQ1,N-cad,E-cad,TGF-beta 1 and the expression of VEGF protein in the relationship with clinical pathological characteristics of ovarian epithelial sex FOXQ1,E-cad,N-cad,TGF-beta 1,VEGF expression is associated with clinical stages,lymph node metastasis(P < 0.05).And has nothing to do with the patients age,organization type,histological grade(P > 0.05).4.FOXQ1,N-cad,cad and TGF-E-beta 1 protein expression in epithelial ovarian cancer in the inspection by the Spearman correlation analysis method,in epithelial ovarian cancer tissues,FOXQ1 and negatively correlated with E-cad protein expression(r = 0.481,P < 0.05),and N-cad in protein expression levels were positively correlated(r = 0.305,P < 0.05).TGF-beta 1 and VEGF protein expression levels were positively correlated(r = 0.531,P < 0.05).Conclusions1.The epithelial ovarian cancer tissues TGF-beta 1,FOXQ1,VEGF and higherN-cad mRNA and protein expression,E-cadmRNA and protein expression decreased,TGF-beta 1 \ FOXQ1 \ E-cadherin \ EMT may,as a new signaling pathways involved in cancer development of epithelial tumors.2.FOXQ1,N-cad,E-cad,TGF-beta 1 and positive expression rate of VEGF in epithelial ovarian cancer tissue is associated with clinical stages,lymph node metastasis.3.FOXQ1 and epithelial ovarian cancer EMT phenomenon is related to,play an important role in the invasion and metastasis of ovarian cancer.
