Cottonseed meal as a feed ingredient is widely used in animal husbandry and production.Gossypol which has reproductive toxicity and other toxic effects is widely found in seed plants.In order to study the mechanism of gossypol on the reproductive toxicity in male mice.Sixty Kunming male mice weighed 20.43±1.57 g were randomly divided into A(control group),B(40 mg/kg),C(80 mg/kg)and D(120 mg/kg),15 mice each group.The mice in 3 dosage groups were treated with gossypol which diluted by 1% sodium carboxymethyl cellulose orally by using gavages needle for 20 days,the control group were given 1% sodium carboxymethyl cellulose.In the 21 st day mice were killed by cervical dislocation and their testis had been obtained.The content of ROS in testis tissue was detected by DCFH-DA probe-notation,the expression of gene mRNA in testis tissue were detested by QRT-PCR,Western blot was used to detect the expression of gene protein in testis tissue.The research result showed that compare with the control group the content of ROS was increased with the increase of gossypol,in the 80,120 mg/kg group were significant increased(P<0.01,P<0.001).the expression quantity of JNK1 mRNA in 40,80,120 mg/kg group were showed a significant rise(P<0.05),the expression quantity of JNK2 mRNA in 40,80,120 mg/kg group were significant increasd(P<0.01),the expression quantity of JNK3 mRNA in 80,120 mg/kg group were significant increasd(P<0.001),the expression quantity of MKK4 mRNA was significant increasd in 80 mg/kg group(P<0.05)and 120 mg/kg group(P<0.001),the expression quantity of FAS and FASL mRNA was significant increasd in 80 and 120 mg/kg group(P<0.01,P<0.001),the expression quantity of C-MYC mRNA in 120 mg/kg group was significant increasd(P<0.001).Moreover in 80 mg/kg group the expression of P-MKK4,P-JNK,FAS and FASL protein were showed a significant rise,and in 120 mg/kg group the expression of P-MKK4,P-JNK,FAS,FASL and C-MYC protein were significant increased.Conclusion,Intragastric administration of 80 and 120 mg/kg of gossypol can significantly affect the activity of ROS-JNK pathway in Kunming male mice testicular tissue.It was showed that gossypol can increase the production of ROS in testis tissue,activate JNK signaling pathway,potentiate the expression of FAS/FASL and C-MYC to induce the apoptosis of testicular cells and damage the reproductive system of male mice.This may be one of the main mechanisms of the gossypol antifertility.
