Objective:Granzyme B secreted by activated immune cells could induce apoptosis of target cell in acute myocardial infarction(AMI).microRNA-874 was identified to target Granzyme B by searching the mi RNA database.The aim of this study is to determine the levels of Granzyme B and microRNA-874 as well as assay their correlations or predict powers in AMI.Method:80 patients with AMI and 40 healthy subjects were enrolled.AMI patients were enrolled from the Department of Cardiology,the Second Hospital of Jilin University from October 20 to January 2017.We measured levels of plasma Granzyme B and microRNA-874 in patients with AMI by ELISA or real-time polymerase chain reaction,The relationship between plasma microRNA-874 and Granzyme B was assessed with the Spearman correlation test.Receiver operating characteristic(ROC)curves were established to evaluate the predictive power of circulating microRNA-874 and Granzyme B for patients with AMI.The area under the ROC curve(AUC)was used to assess the predictive power and the best cut-off point for the selected variables.Results:1?In this study,we also measured the plasma level of Granzyme B in AMI patients and health subjects.The result showed that plasma level of Granzyme B was significantly increased in AMI patients(16.71±7.23ng/ml),compared with control(9.27±3.90 ng/ml)(p< 0.01).2?We also assayed the plasma level of microRNA-874 in AMI patients and health subjects.The result showed that plasma level of microRNA-874 was significantly decreased in AMI patients(0.20±0.17 fold changes),compared with control(1.00±0.79 fold)(p< 0.01).3?We further assay the correlation of Granzyme B and microRNA-874.The results showed that microRNA-874 was negatively correlated with Granzyme B(R =-0.237;p < 0.05)?4?We performed ROC analysis to evaluate the predictive power of circulating Granzyme B or microRNA-874 for AMI.the AUC for Granzyme B was 0.829(95% CI: 0.749–0.909;p < 0.01)and the optimal cut-off value was 11.44 with sensitivity and specificity of 73.8 and 70.0%,respectively.microRNA-874 was significantly decreased;we used 1/ microRNA-874 to predict the AMI.The AUC for microRNA-874 was 0.815(95% CI:0.743–0.892;p < 0.01)and the optimal cut-off value was 0.1576 with sensitivity and specificity of 80.0 and 70.9% respectively.We further corroborated the discriminatory value of Granzyme B and microRNA-874 by binary logistic regression analysis.Similar prognostic properties of Granzyme B(odds ratio: 1.304;95% CI: 1.168–1.445;p < 0.01)and microRNA-874(odds ratio: 0.068;95% CI: 0.022–0.209;p < 0.01)for AMI.Conclusion:1,We found that plasma Granzyme B was significantly higher and microRNA-874 was significantly lower in AMI patients,and there was a negative correlation of microRNA-874 with Granzyme B,indicating microRNA-874 might target Granzyme B2,Plasma microRNA-874 and Granzyme B can be used as a marker for the diagnosis of AMI.
