Objective:To study the effect of Bai Hu Ren Shen Decoction on the oxidative stress level of TAOC,the pathological state of sciatic nerve and the expression of TRPV1 mRNA in DRG rats.So as to provide experimental basis and new target for clinical treatment of diabetes mellitus and its complications.Methods:Forty-five healthy male Sprague-Dawley(SD)rats were randomly divided into 5 cages and cultured for 1 week.After 12 hours of fasting in rabbits,diabetic rats were established by intraperitoneal injection of streptozotocin 50 mg/kg.Blood glucose ?16.7 mmol/L was used as a standard for diabetic rats after 72 hours of STZ injection.The rats were randomly divided into model group,Bai Hu Ren Shen Decoction group and?-lipoic acid group.Bai Hu Ren Shen Decoction to 9.38g/kg/d gavage,?-lipoic acid group was given 20mg/kg/d intraperitoneal injection,The model control group and the normal control group were given normal saline.Observe the general state of each group of rats.After 8 weeks of intragastric administration,blood samples were taken after anesthesia.The TAOC and C-P were detected by ELISA.Electron microscopy was used to observe the sciatic nerve pathology of each group,The expression of TRPV1 mRNA in the dorsal root ganglion was detected by RT-PCR.Results:1.C-peptide:Compared with the normal group,the content of C-peptide in the model group and the two treatment groups was significantly lower than that in the control group(P<0.01).Compared with the model group,the content of C-peptide in the group(P<0.05),while the Bai Hu Ren Shen Decoction group and a-lipoic acid group was no statistically significant(P>0.05).2.TAOC:Compared with the normal group,the total antioxidant capacity of the model group,the Bai Hu Ren Shen Decoction group and the a-lipoic acid group was significantly lower than that of the normal group(P<0.05),compared with the model group,(P<0.05).Compared with the a-lipoic acid group,the total antioxidant capacity of the group was significantly higher than that of the a-lipoic acid group(P<0.05).3.TRPV1 mRNA:Compared with the normal group,the expression of TRPV1 mRNA in the dorsal root ganglion of the model group,Bai Hu Ren Shen Decoction group and the a-lipoic acid group was significantly lower than that in the normal group(P<0.01).Compared with the model group,(P<0.01).There was no significant difference between the two groups(P>0.05).The expression of TRPV1 mRNA in the dorsal root ganglion of the a-lipoic acid group was significantly higher than that in the a-lipoic acid group(P>0.05).4.Sciatic nerve under electron microscopy:In the normal control group,the sciatic nerve of the sciatic nerve was clear and intact.The myelin sheath was arranged in concentric circles,showing complete cells and internal structures such as mitochondria and nuclei.The sciatic nerve myelin was significantly reduced and degenerated in the model group.Partial lamellar separation,Schwann cell nuclear pyknosis and inflammatory cell infiltration were observed.The two treatment groups improved compared with the model group.Conclusion:1.Bai Hu Ren Shen Decoction can improve improve the C peptide,with the function of protecting islet ? cells.2.Bai Hu Ren Shen Decoction can improve the level of serum TAOC in diabetic rats,alleviate the oxidative stress state of diabetic rats,and have good antioxidant energy.3.Bai Hu Ren Shen Decoction can down-regulate the expression of TRPV1 mRNA in DRG.4.Bai Hu Ren Shen Decoction can improve the damage of sciatic nerve by high glucose.
