Objective:By observing the effect of electro acupuncture at different frequency on the expression of uncoupling protein 5(UCP5)in the ischemic area of cerebral infarction rats,to explore the central mechanism of electroacupuncture treatment of cerebral infarction.Methods:Sixty healthy male SD rats were randomly divided into blank group,sham operation group,mo del group,2 Hz electroacupuncture group,50Hz electroacupuncture group and 100Hz electroacupuncture group,each group of 10 rats.Model group and different frequency electroacupuncture groups were used to prepare the middle cerebral artery occlusion(MCAO)model.Rats in the blank group,sham operation group and model group were not treated,only the binding treatment was performed.Electro acupuncture groups were taken the rat ipsilateral "El 10" and "SJ 5" with the continuous wave of 2Hz,50Hz and 100Hz,once a day for 20 minutes,continuous treatment for 21 days.Each group of rats were subjected to a screen test on the 1st,3rd,7th,14th and 21st day after the operation,to detect the neurological deficits in rats.Rats in each group were sacrificed on the right side of the cerebral cortex on the 21st day,and the positive expression of UCP5 in cerebral ischemic region was detected by the immunohistochemistry.At the end of the study,SPSS20.0 statistical software was used to analyze the data.Results:1.Behavioral scoring results was shown:2Hz group compared with the model group,50Hz group,100Hz group:the motor function of the 2Hz group was better than that of the model group at the 21st day after operation,the difference was statistically significant(P<0.05).50Hz group compared with the model group,2Hz group,100Hz group:compared with the model group,the motor function of the 50Hz group was better than that of the model group at the 7th day after operation,the difference was statistically significant(P<0.01);compared with the other groups,the motor function of the 50Hz group was better than that of the other groups at the 14th day after operation,the difference was statisti-cally significant(P<0.05);compared with the other groups,the motor function of the 50Hz group was better than that of the other groups at the 21st day after operation,the difference was statistically signifi-cant(P<0.01).2.The expression of UCP5 in the cerebral cortex of rats was shown:There was no significant difference between the blank group and the sham operation group,the 100Hz group(P>0.05);There was no significant difference between the model group and the electroacupu-ncture groups(P>0.05);Compared with the blank group,the brain protection of the model group was better than that of the blank group,the difference was statistically significant(P<0.01).Different frequency EA groups compared with the other groups:?2Hz group compared with the blank group,50Hz group,100Hz group:compared with the blank group,the brain protection of the 2Hz group was better than that of the blank group,the difference was statistically significant(P<0.01);There was no significant difference between the 2Hz group and the 50Hz group(P>0.05);2Hz group compared with the 100Hz group,the brain protection of the 2Hz group was better than that of the 100Hz group,the difference was statistically significant(P<0.05).?50Hz group compared with the blank group,100Hz group:compared with the blank group,the brain protection of the 50Hz group was better than that of the blank group,the difference was statistically significant(P<0.01);50Hz group compared with the 100Hz group,the difference was not statistically significant(P>0.05).Conclusion:1.Electroacupuncture can improve the motor nerve dysfunction in rats with cerebral infarction and promote the recovery of its upper limbs motor function.Electroacupuncture at different frequency can promote the recovery of upper limbs,of which 50Hz EA group the most significant effect,followed by 2Hz and 100Hz EA group.2.Electroacupuncture treatment has brain protection on cerebral infarction,and its mechanism may be related to the up-regulation of UCP5 in ischemic area.The electroacupuncture at different frequency can up-regulate the expression of UCP5.The effect of 2Hz EA group is the most significant,followed by 50Hz and 100Hz EA group.
