Objective:To investigate the mechanism of scalp electroacupuncture(EA)in the treatment of cerebral infarction in rats,the effects of scalp EA of different frequencies on the motor function and the expression of neureglin1(NRG-1)in brain were observed.Methods:60 Sprague Dawley rats were randomly divided into 6 groups: blank group,sham-operated group,model group,2Hz,50 Hz and 100 Hz EA group.The middle cerebral artery occlusion(MCAO)model was made in the model group and the EA group by the method of thread occlusion.In the sham-operated group,only tissue detachment and vascular separation were performed,without thread occlusion.In the EA group,Baihui(GV20)and Qianshencong(the one at the front of EX-HN1)were pricked to the diseased side,the depth of acupuncture was about 0.5 inch,the stimulation frequency was 2Hz,50 Hz and 100 Hz respectively,the treatment time was 20 minutes each time,once a day,and the treatment period was 14 days.The rats in the blank group,the sham-operated group and the model group were not treated with EA,but only with the same binding as that in the EA group.On the first,third,fifth,seventh and fourteenth day of the treatment,the rats in each group were scored for the traction test of the forelimb grasp to detect the motor function.At the end of the last treatment,the brain tissues of each group were perfused and collected.Enzyme linked immunosorbent assay(ELISA)was used to detect the expression of NRG-1 in the right cerebral tissue of rats,immunohistochemistry was used to observe the positive expression of NRG-1in the right cortical ischemic area of rats,and the correlation between behavioral score and NRG-1 expression was analyzed.The experimental data were analyzed by spss23.0.Results:1.The results of behavioral scores showed that:In the process of making model,the operation of peeling tissue and separating blood vessels will not affect the motor function of rats,there was no significant difference in motor function between the blank group and the sham-operated group,The difference was not statistically significant(P>0.05).After modeling,the motor function of rats decreased in different degrees,the difference was statistically significant(P<0.05).EA can promote the recovery of motor function of rats.On the 14 th day of treatment,the motor function of each EA group was better than that of the model group,the difference was statistically significant(P<0.01).The recovery of motor function in 2Hz EA group was the best,and its score was close to that in the blank group,with no significant difference(P>0.05).From the view of the time of treatment,from the 7th day of treatment,the motor function of rats in each EA group improved significantly.On the7 th and 14 th day of the treatment,the motor function scores of each EA group were significantly higher than that of the model group,the difference was statistically significant(P<0.01).From the view of EA frequency,the improvement of motor function of rats by EA of different frequencies is different.On the 5th and 14 th day of treatment,the therapeutic effect of EA group is different.The motor function of rats in 2Hz EA group is better than that in 50 Hz EA group and 100 Hz EA group,the difference is statistically significant(P<0.05);there is no difference in therapeutic effect between 50 Hz EA group and 100 Hz EA group,and the difference is not statistically significant(P>0.05))2.The results of qualitative and quantitative analysis of NRG-1expression in brain tissue in rats showed that:Immunohistochemical localization and qualitative analysis showed that there were a small number of NRG-1 positive cells in the normal brain tissue of rats,and cerebral infarction as a stimulating factor could up regulate the number of NRG-1 positive cells in ischemic penumbra.The expression rate of NRG-1 positive cells was different in different frequency EA.The AOD valueof 2Hz EA group was higher than that of model group,50 Hz and 100 Hz EA group,the difference was statistically significant(P<0.05).The results of ELISA showed that the effect of EA at different frequencies on the expression of NRG-1 was different,and the effect of 2Hz EA group was the best,the expression of NRG-1 was higher than that of model group,50 Hz and 100 Hz EA group,the difference was statistically significant(P<0.05).3.The analysis of correlation between motor function score and NRG-1expression in rats showed that:There was a positive correlation between the score of motor function and the expression of NRG-1 in 2Hz EA group,the difference was statistically significant(P<0.01).It is suggested that the mechanism of EA in promoting the recovery of motor function in rats with cerebral infarction may be related to the up-regulation of NRG-1 expression in the ischemic area.Conclusion:1.Scalp EA with different frequency can improve the motor function of forelimb and promote the expression of NRG-1 in the ischemic area of rats with cerebral infarction.2.The mechanism of EA on the head in the treatment of cerebral infarction may be related to the up regulation of NRG-1 expression in the ischemic area to play the role of brain protection such as inhibit the apoptosis and inflammatory response.
