Background: Breast cancer is the most common malignant tumor of female,which causes serious damage to women's health and life.It is of great significance to explore novel tumor-specific markers for early diagnosis and treatment of breast cancer.Present studies show that high expression of TYK2(Tyrosine Kinase 2)can promote proliferation and invasion of tumor cells.However,its role in the development of breast cancer has cell and tissue specificity,which needs further research.P53,known as famous tumor suppressor gene,plays important part in regulating cell cycle,apoptosis,and so on.Nevertheless,P53 mutation in tumor could promote malignant development.Recently a study showed that induction of wild-type P53 expression in lung cancer cells could downregulate the expression of TYK2 through an E3 ubiquitin ligase SIAH2(seven-in-absentia-2),which would affect downstream signaling pathways.This showed that there is regulating relationship between P53 and TYK2.However,the correlation between the two genes in breast cancer and corresponding clinical significance are still poorly understood.Objective: The objective of our study is to investigate TYK2 and P53 expression in breast cancer,as well as analyze the relationships between gene expression and clinical pathology features.We further demonstrate whether there is correlation between TYK2 and P53 expression in breast cancer,and elucidate corresponding clinical significance.Method: 102 breast cancer,30 fibroadenoma and 20 para-carcinoma tissues were achieved from the Second Affilited Hosptital of Dalian Medical University from September,2013 to September,2016.Immunohistochemical staining method was used to detect the expression of TYK2 and P53 in three different tissues.We analyzed the relationships between gene expression and clinical pathology features,as well as correlation between two genes.Results:(1)TYK2 is highly expressed in breast cancer tissues,the positive rate is 57.84%(59/102),which is higher than that of fiberadenomas(6.67%)and para-carcinoma tissues(0.00%).P53 is also highly expressed in breast cancer tissues,the positive rate is 40.20%(41/102),higher than that of fiberadenomas(3.33%)and para-carcinoma tissues(0.00%).(2)TYK2 expression in breast cancer tissues is related to tumor size,clinical stage,ER,PR and Ki-67 status(p<0.05).However,there are rare relationships among TYK2 expression and age,menopause status,lymph node metastasis,vascular or nerve invasion,HER-2 status(p>0.05).P53 expression in breast cancer is related to lymph node metastasis,ER,PR and Ki-67 status(p<0.05).Nevertheless,there are rare relationships among P53 expression and age,menopausal status,tumor size,vascular or nerve invasion,clinical stage,HER-2 status(p>0.05).The expression rates of TYK2 in molecular subtypes of breast cancer are 42.31%(Luminal A),54.55(Luminal B),61.90%(HER-2-overexpression type)and 77.27%(triple negative type);the expression rates of P53 are 19.23%(Luminal A),33.33%(Luminal B),42.86%(HER-2-overexpression type)and 72.73%(triple negative type).P53 expression is the highest in triple negative breast cancer,with a statistically significant difference(X2=15.148,p=0.002).(3)The expression of TYK2 and P53 in the overall population of breast cancer is significantly correlated(r=0.560,p=0.000).Conclusion:(1)TYK2 and P53 are highly expressed in breast cancer,their expression rates are higher than those of fiberadenomas and para-carcinoma tissues;P53 expression level is high in triple negative breast cancer.(2)TYK2 expression in breast cancer tissues is related to tumor size,clinical stage,ER,PR and Ki-67 status.P53 expression is related to lymph node metastasis,ER,PR and Ki-67 status.(3)TYK2 expression is positively correlated with P53 in breast cancer;detection of both TYK2 and P53 is of great significance during diagnosis,treatment and prognosis evaluation of breast cancer.
