The Expression Of LncRNA NONHSAT040475 In Acute Graft-versus-host Disease

Background: Acute graft-versus-host disease (aGVHD) is a vital factor that affects the overall curative effect of allogeneic hematopoietic stem cell transplantation. At present, the diagnose of aGVHD mainly depends on clinical manifestations. But due to some cases of clinical manifestation is not typical, and clinical doctors' subjective factors, the diagnose of aGVHD is still not perfect. Therefore, looking for the objective and specific diagnostic biomarkers is particularly important for early warning, diagnosis, treatment and prognosis of aGVHD. Our previous study and preliminary experiment found that the expression of Inc RNA NONHSAT040475 was high in cGVHD patients compared with non GVHD patients and healthy controls. So we explore further the relationship between the expression of Inc RNA NONHSAT040475 and aGVHD.Objective: This study aims to explore the expression change of lncRNA NONHSAT040475 in aGVHD patients and its clinical significance. And then find objective and specific biomarkers and lay the foundation of aGVHD warning, diagnosis and developing target drugs.Methods:(1) Collect fresh peripheral blood in patients within 100 days after allo-HSCT including 23 aGVHD patients, 24 non aGVHD, and another 22 healthy donors. Samples of patients in different time points and patients with engraftment syndromes, intestinal infection,liver dysfunction and drug rush were also collected.(2) Statistics into the clinical characteristics of cases in groups including age, sex,transplantation type, regime type and disease and analyze the statistical significance.(3) Extract the mononuclear cells of samples in three groups, then use qPCR to detect the different expression of lncRNA NONHSAT040475 in patients with aGVHD, non aGVHD and healthy volunteers.(4) Use qPCR to detect the expression changes of lncRNANONHSAT040475 in patients in different time points.(5) Use qPCR to detect the expression changes of IncRNA NONHSAT040475 in patients with engraftment syndromes, intestinal infection, liver dysfunction and drug rush.Results:(1) There is no statistically significant differences between aGVHD and non aGVHD patients in age, sex, transplantation type, regime type and disease.(2) Compared with the control group, the expression of IncRNA NONHSAT040475 is significantly increased in aGVHD group (P < 0.01).(3) The expression level decreased significantly in aGVHD patients after treatment (P <0.05).(4) There seems no obvious correlation between the expression level of IncRNA.and aGVHD severity classification.(5) Compared with aGVHD, the expression of IncRNA is low in patients with other complications after transplantation (P<0.05).(6) There is an apparent increased tendency of IncRNA before the onset of aGVHD.Conclusions:(1) LncRNA NONHSAT040475 may provide a method for diagnosis and differential diagnosis of aGVHD.(2) The expression of IncRNA NONHSAT040475 cannot reflect the severity of aGVHD.(3) The dynamic expression of patients after allo-HSCT indicates that IncRNA NONSAT040475 may provide new clues to aGVHD warning.
