We used BGC823/Rab27A as experimental group, and BGC823/Vector as control group. MTT, soft agar colony formation and transwell assay were used to observe the malignant behavior.Results: The migrative and invasive ability of BGC823/Rab27A cells were obviously reduced after Rab27A overexpression, but there was no difference on the proliferation of BGC823/Rab27A cell line.Conclusion: Rab27A might be an anti-oncogene during the development of gastric cancer,inhibiting the migrative and invasive ability of gastric cancer cells, which will provide new theory evidence and potential therapeutic target to the clinical treatment of gastric cancer.Objective: To study the influence of Rab27A protein derived from colon carcinoma cells on the cytotoxicity of NK cells against cancer cells by regulating the secretion of exosome and soluble molecules.Methods: Through previous study, Rab27A was low expressed in colon carcinoma cell line RKO, while overexpressed in SW480. The secretory volumes of exosome relatively coming from RKO cells and SW480 cells was detected by ELISA and BCA. MTT was used to observe the proliferation rate of RKO cells and SW480 cells. With CCK-8, the cytotoxicity of NK cells against RKO and SW480 cells was examined, and the influence of exosome and soluble molecules secreted by RKO cells to the cytotoxicity of NK cells against cancer cells was observed.Results: The secretory volume of exosome from RKO cells and the cytotoxicity of NK cells against RKO cells were obviously lower than SW480 cells, but the proliferation rate of RKO cells was apparently faster than SW480 cells. exosomeand soluble molecules secreted by RKO cells reduced the cytotoxicity of NK cells against cancer cells.Conclusion: exosome and soluble molecules secreted by RKO colon carcinoma cells could evidently inhibit the cytotoxicity of NK cells against cancer cells. | | Keywords/Search Tags: | Rab27A, gastric cancer, proliferation, migration, invasion, exosome, cytotoxicity, colon carcinoma, proliferation rate, soluble molecules | PDF Full Text Request | Related items |
