Iron,a trace element,is indispensable to the human body.It is involved in many important physiological and biochemical reactions in the human body,such as ATP synthesis,cell respiration,cell proliferation,electron transfer,hemoglobin synthesis,DNA self-replication and regulation.Changes in the content of trace iron in the human body will be associated with some diseases.The lack of trace iron in the human body can lead to anemia,physical weakness,fatigue,cell normal function decline,decreased protease activity and normal tissue abnormalities.When the trace iron overload in the human body,it can also cause a series of diseases,such as osteoporosis,Alzheimer's disease,Parkinson's disease,arteriosclerosis and so on.The absorption,intake,utilization and reserve of trace iron are regulated by precision mechanism,but there is no effective mechanism for the excretion of excess trace iron in human body.So when too much exogenous trace iron is taken in which would result in iron overload.Copper,another trace element only less than iron,is also indispensable to the human body.It is very important in the normal growth and development of the human body.It plays an irreplaceable role in the bones,nervous system,hematopoietic system mature.Similar to trace iron,abnormal metabolism of trace copper can also cause many diseases,such as immune deficiency,hyperlipidemia,coronary heart disease,osteoporosis and cancer.The absorption,intake,utilization and reserve of trace copper are regulated by precision mechanism in human body,but there is no effective mechanism for the excretion of excess trace copper.When too much exogenous trace copper was taken in which would result in copper overload.There are a large number of literatures and clinic reports showing that some prescriptions of the traditional Chinese medicine(TCM)can alleviate or cure disease caused by iron or copper overload.But the mechanism of TCM for treatment of these diseases caused by iron or copper overload is not clear.It is speculated that some complex agents existed in the decoction of TCM,which form complex with iron or copper and excreted.In order to confirm the speculation and assess the complex ability of TCM for iron and copper,a chemical screen method was established in this laboratory previous.In this dissertation,a cytologic method was established to assess the complex ability of TCM for iron and copper.The MTT method was used to study the effect of TCM on the differentiation of MC3T3-E1 and HepG2 cells.The model of iron and copper overload was set up,the different TCM were used to intervene,so as to evaluate the complex ability of TCM to iron ion and copper.The results showed that TCM with the complex ability to iron over 70%are fructus alpiniae oxyphyllae,fructus psoraleae,cynomorium songaricum,flatstem milkvetch seed,jujube kernel,cistanche,medicinal cyathula root,leonurus,Salvia miltiorrhiza,pseudo-ginseng and ephedra.The highest rate of complex ability was 92.42%for cyathula officinalis and 88.65%forfructus psoraleae.The TCM with complex ability to copper over 70%are china rose,tree peony bark,chinese gall,pomegranate rind,syzygium aromaticum.The highest complex ability was 94.65%for pomegranate rindand 92.78%forchina rose.The results of complex ability of TCM to iron and copper by MTT are agreement with those by chemical methods previous in this group.
