Objective Chronic prostatitis?CP?has been a common and recurrent disease in male.For its high incidence and low remission rate,CP can lead to sexual dysfunction and male infertility.Etiological research of CP have been focused on pathogenic microorganism,abnormal function of epithelial cells and urine reflux.However,the pathogenesis of CP remain unclear.In recent years,correlational studies have demonstrated that there is a relationship between cytokines and neuroendocrine and CP.To explore difference of intracellular calcium ion([Ca2+]i)concentration of nerve growth factor?NGF?and chromogranin A?CgA?treated prostate epithelial cell line RWPE-1 cells and evaluate the role of N GF and CgA in CP.Methods Using different concentrations of NGF and C g A?50ng/mL and 200ng/mL?to stimulate RWPE-1 cells,the difference of intracellular [Ca2+]i concentration were observed by using Laser Scanning Confocal Microscopy?LSCM?.The difference of intracellular[Ca2+]i concentration were observed after infusing NGF and CgA mixed with TRPV1 receptor antagonist.When the extracellular solution was changed to DPBS,the intracellular [Ca2+]i concentration were observed after N GF and CgA infusion.Results Change in intracellular [Ca2+]i concentration did not occur by infusing 50ng/mL NGF and C g A.Intracellular [Ca2+]i concentration significant increased by infusing 200ng/mL NGF and CgA.Intracellular [Ca2+]i concentration did not change after infusion of NGF and C g A mixed with TRPV1 receptor antagonis t.Even when the extracellular solution was changed to D-PBS,the intracellular [Ca2+]i concentration did not change after NGF and C g A infusion.Conclusions NGF and CgA induced increases in intracellular [Ca2+]i depend on the opening of TRPV1,then extracellular calcium ions enter the cells.Thus,NGF and C g A might participate in the mechanism of CP and caused the opening of TRPV1 of prostate epithelial cell membrane.Then,the intracellular [Ca2+]i concentration was increased.In the end,prostate epithelial secretion was declined.
