Objective Establish the model of Autism Spectrum Disorder with exposuring to Poly –IC,Analysis the change of development,behavior,and nervous pathological,to clear if the Sprague-Dawley rats is suitable for the study of autism spectrum disorders.Methods Ten sprague-dawley rats are randomly divided in two groups of each 5.The rat Poly–IC-induced autism model is injected intraperitoneally with 5mg/kg Poly–IC at gestational day 12,while the control group rats are injected intraperitoneally with 9g/L normal saline.Record each offspring rat body weight,negative geotropism,open eye time,swimming,morris water maze,social communication ability test,evaluation of rat development situation and the existence of antism-like social behavior;The pathological changes were observed in hippocampus and prefrontal cortex of 48 day old rats by immunohistochemical staining and indirect immunofluorescence staining.Results Compared with control group,each measuring point,experimental rat weight is lighter(experimental control:13.23±1.34,19.97±1.18,29.74±0.86,166.97±8.35,281.9±11.21;control group:13.75±1.23,29.06±1.33,40.10±0.77,180.9±10.22,298.82±7.12,),open eye time delay(experimental control:0.00±0.00,0.00±0.00,0.80±0.42,1.50±0.53,control group:0.60±0.51,1.30±0.48,1.50±0.53,2.00±0.00),swimming score is low(experimental control:0.70±0.48,1.00±0.47,2.00±0.67,3.10±0.57,control group:2.60±0.70,3.00±0.00,3.70±0.48,3.70±0.48),the above differences are significant(P < 0.05);Negative hasten to sexual sensitivity decreased,but only P10,there is significant difference(experimental control:11.05±2.03,control group:7.56±1.58,P < 0.05),there was no significant difference of the other points."Near-escape" scores in social communication ability test on the low side(experimental control:-87.50±56.92,control group:81.89±70.93).The incubation period extended obviously of the Morris water maze experiment(experimental control:92.68±24.81,85.50±25.03,69.78±24.68,60.88±27.99,control group:61.75±21.46,40.45±18.00,40.78±30.28,21.55±11.10),right across the platform number decreased significantly(experimental control:2.20±1.32,control group:6.10±2.47),and there were significant difference(P<0.05).The results of immunohistochemical staining showed that the experimental group rat frontal cortex astrocytes glial cell proliferation,enhanced staining,control group rat frontal cortex hyperplasia is not obvious,weakly positive staining Indirect immunofluorescence results showed that the experimental group rat hippocampal CA1 area MR average optical density value(0.0613±0.0288)and the GR mean optical density value(0.0419±0.0403)than the control group(MR:0.0813±0.0539;GR: 0.0612±0.0436),with significant difference(t=10.319,10.241;P <0.05);Experimental group MR/GR ratio(the same vision area under MR and GR optical density value ratio)and the control group there was no significant difference.Conclusion The exposure with Poly-IC during early pregnancy can cause motor development lags behind,harm the social communication ability,social communication ability is impaired,decreased memory learning abilities,sample ASD symptoms of Sprague-Dawley rat,that maybe associated with the activation of astrocytes and the abnormal expression of MR and GR on the surface of hippocampal cells,hence Sprague-Dawley rats pregnant early Poly-IC exposure can be used for the establishment of the model of autism spectrum disorder,provide a platform for the research of autism spectrum disorders.Hence exposure with Poly-IC during early pregnancy can be used for the establishment of the model of autism spectrum disorder,provide a platform for the research of autism spectrum disorders.
