Objective This study aims to detect the expression of RASSF1A(The Ras-Association Domain Family 1 isoform A)gene in breast cancer tissues and corresponding adjacent tissues,analyze the correlation between the expression of RASSF1 A gene and the clinical pathological characteristics and explore the possible clinical significance of RASSF1 A gene.Methods 1.60 cases of breast cancer tissues and corresponding adjacent tissues which were confirmed by definite pathological diagnosis were collected,fixed with formalin and made into paraffin sections.The expression of RASSF1 A gene in 120 specimens was detected by immunohistochemical staining.2.The clinical and pathological data of 60 cases were collected,including gender,age,maximum tumor diameter,lymph node metastasis,tumor histological grade,vascular invasion,ER,PR,Her-2,Ki-67 and clinical TNM staging.3.SPSS statistical analysis software was used to analyze the correlation between the expression of RASSF1 A gene and clinical and pathological data.Result 1.The immunohistochemical staining showed that the positive expression rate of RASSF1 A gene in cancer tissue was 35%(21/60),and the positive expression rate of RASSF1 A gene in adjacent tissues was 86.7%(52/60).The positive expression rate of RASSF1 A gene in cancer tissue was significantly higher than that in breast cancer tissue(P <0.05).2.Statistical analysis indicated that the expression of RASSF1 A gene is correlative with lymph node metastasis(P=0.004),histological grade(P=0.011),vascular invasion(P=0.038)and Her-2(P=0.002),Ki-67(P=0.011).However,there was no significant correlation between the expression of RASSF1 A gene with gender(P>0.99),age(P=0.740),maximum tumor diameter(P=0.585),number of lymph node metastasis(P =0.087),ER(P =0.681)and PR(P=0.597).3.The clinical TNM staging of 31 patients was locally early(stage I,II,and stage III A(T3N1MO)),and the positive expression rate of RASSF1 A gene was 48.4%(15/31).The clinical TNM staging of 29 patients was locally advanced(stage III(except T3N1MO)),and the positive expression rate of RASSF1 A was 20.7%(6/29).The positive expression rate of RASSF1 A protein in locally early breast cancer patients was higher than that in patients with locally advanced stage(P =0.025).Conclusion 1.The expression of RASSF1 A gene may be involved in the occurrence and development of breast cancer.2.RASSF1 A gene may be a better marker for prognosis of breast cancer patients.
