background and Objective: Gastric cancer stem cells is a hot research topic in recent years.The proliferation and differentiation of gastric cancer stem cells are closely related to the metastasis and prognosis of gastric cancer ?HDAC1 play an important role on proliferation and differentiation in embryonic stem cell?Effect and mechanism of HDAC1 on proliferation and differentiation in Gastric cancer stem cells have not been reported?To explore the effect and mechanism of HDAC1 expression on the proliferation and differentiation in human gastric cancer stem cells(GCSCs),in this study,we observe the effect of HDAC1 expression on the proliferation?immigration and invasion and detect the expression of stemness-related genes in human gastric cancer stem cells which were transfected by lentiviral vector ?Methods: The GCSCs were selected by flow cytometry with Nanog as marker.The expression of HDAC1 in GCSCs was Down-regulated by lentiviral vector and detected by real-time quantitative reverse transcription PCR(q RT-PCR)and Western Blot.The difference of Proliferation and immigration between two groups were evaluated by MTT assays and scratch test respectively;The difference of invasion between two groups were evaluated by transwell assays;The expression of stemness-related genes was detected by qRT-PCR and Western Blot.Results: The expression of HDAC1 was obviously down-regulated in the experimental group(p<0.05)?The proliferation and immigration in the experimental group were obviously lower than control group(p<0.05).The expression of stemness-related genes in the experimental group were obviously lower than control group(p<0.05).Conclusion: The low expression of HDAC1 can decrease proliferation?differentiation and invasion in gastric cancer stem cells?The low expression of HDAC1 down-regulate the expression of BMI1,which was thought to be one of the mechanism that HDAC1affect proliferation and differentiation in human gastric cancer stem cells.
