Objective To observe the inhibitory effect of curcumin on corneal neovascularization(CNV)after alkali burn in rabbit corneal,and to explore its possible mechanism.Provide a theoretical foundation for clinical application in the future.Methods The alkali burn models were established by placing a filter paper saturated with 1mol/L Na OH at the central corneas of right eyes of 48 rabbits for 1 minute,which were distributed randomly into 4 groups: group A dimethyl sulfoxide(DMSO)control group,group B 40 ?mol / L curcumin group,group C 80 ?mol / L curcumin group and group D 160 ?mol / L curcumin group,twelve rabbits(12 eyes)were in each group.Right eye points corresponding to the drug,1 drop / time,4 times / day,continuous medication for 14 days.(1)The corneal neovascularization was observed by the slit lamp and the rabbit cornea was photographed 2 days,4 days,7 days and 14 days after alkali burn.The CNV length was calculated and the CNV area was calculated.(2)Three rats were randomly selected from group A,group B,group C and group D for 2 days,4 days,7 days and 14 days after alkali burn,the aqueous humor was obtained and using enzyme-linked immunoassay(ELISA)to detect VEGF,4 ebp1 factor of protein content in the aqueous humor;Three rabbits were randomly selected from group A,group B,group C and group D for 7 days and 14 days after alkali burn,the cornea was obtained and using reverse transcription polymerase chain reaction(RT-PCR)to detect 4EBP1,P70S6 K,m TOR factor gene(m RNA)in relative expression corneal tissue.(3)The statistical analysis was performed using SPSS23.0 software.The statistical description of the measurement data was expressed by the mean ± standard deviation,and the single factor analysis of variance showed that P <0.05 was statistically significant.Results(1)2 days after cornea alkali burn,the four groups were showed conjunctival hyperemia,alkali burn area cornea gray edema,edema after iris texture obscure,corneal limbus vasodilation congestion,no corneal neovascularization;4 days after cornea alkali burned,the four groups of conjunctival hyperemia began to reduce,the burn area edema,peripheral cornea can be seen new blood vessels grow into.There was no significant difference in the average area of corneal neovascularization between the experimental group(group B,group C,group D)and control group(group A)(P> 0.05);7 days after cornea alkali burned,the corneal neovascularization was strong,thrive and lumen bulky in the control group(group A).The conjunctival hyperemia was significantly reduced in the experimental group(group B,group C,group D)compared with the control group.The new blood vessels invaded the cornea from the limbus,but the growth was slower than that of the control group.There was significant difference in the average area of corneal neovascularization between the experimental group(group B,group C,group D)and the control group(group A)(P <0.05);14 days after cornea alkali burned,Control group(group A)new blood vessels dense thick,net-like the entire cornea;the experimental group(group B,group C,group D)neovascularization was infused into the burn area around the cornea and grew slowly,which was significantly shorter and sparse compared with the control group(group A);The experimental group(group B,group C,group D)compared with control group(group A)average corneal neovascularization area difference was statistically significant(P < 0.05)and with the increase of the concentration of curcumin,the average of corneal neovascularization is reduced gradually.(2)Rabbit corneal alkali burn after 7 days,14 days,the experimental group(group B,group C,group D)compared with control group(group A),amount of 4 ebp1,the expression of VEGF protein in the aqueous humor differences statistically significant(P < 0.05),increased with the concentration of curcumin VEGF,4 ebp1 protein expression quantity reduced in turn.Line Pearson correlation analysis found that 4 ebp1 in aqueous humor is linear positive correlation with VEGF protein expression.Correlation coefficient r of 0.969 in group A and group B is 0.803,0.67,group C is 0.67,group D was 0.972(at the 0.05 level,correlation significantly).(3)Rabbit corneal alkali burn after 7 days,14 days,the experimental group(group B,group C,group D)compared with control group(group A),corneal tissue m TOR,4 ebp1,P70S6 K in the expression of m RNA level difference was statistically significant(P < 0.05),and increased with the concentration of curcumin m TOR,4 ebp1,P70S6 K in the expression of m RNA level quantity reduced in turn.Conclusion 1.Curcumin can inhibit the growth of CNV in corneal alkali burned rabbits.2.With the increase of curcumin concentration,the expression of VEGF protein in aqueous humor decreased;3.Curcumin may reduce VEGF expression by inhibiting m TOR signaling pathways.
