Objective The aim of present study is to analyze serum level of IL-4 and IL-13 in SSc,and investigate its relationship with SSc clinical features.Methods The study included 60 SSc patients and 60 healthy controls with a casecontrol study design.All cases were recruited from the department of Rheumatology and Immunology inpatient and outpatients in the Affiliated Hospitals to Anhui Medical University.The diagnosis criteria of SSc use the American College of Rheumatology in1980 criteria.Self-designed questioner was used to collect the relevant data about SSc patients,and at the same time,5 ml peripheral blood was collected from patients and controls.Specific enzyme linked immunosorbent assay(ELISA)kits were used to detect the serum level of IL-4 and IL-13.t test or Mann-Whitney U test was used for data analysis in SPSS16.0 software.Test level was 0.05.Results The mean serum level of IL-4 in SSc patients was 3.180(2.413,3.188)pg/ml,and its in healthy controls was 2.891(1.999,3.441)pg/ml,there was no difference between serum level of IL-4 in SSc patients and healthy controls.The mean serum level of IL-13 in SSc patients was 3.735(2.951,4.459)pg/ml,significantly higher than that in healthy controls(2.707 [1.553,3.130] pg/ml)(Z=-5.134,P<0.001).A significant association between serum IL-4 level and disease duration and skin thickening have been observed in SSc(?2=7.742,P=0.021;Z=-2.092,P=0.036).Conclusions Serum level of IL-13 in SSc patients was higher than that in healthy controls.There was no difference between SSc patients and healthy controls in serum level of IL-4,but serum level of IL-4 in SSc might relate to disease duration and skin thickening.Aim We aimed to investigate the role of IL-4-590C/T(rs2243250),IL-4R-Q576R(rs1801275),IL-13-1112C/T(rs1800925)and IL-13RA1-43163G/A(rs6646259)single nucleotide polymorphism in systemic sclerosis.Methods The study included 95 SSc patients and 95 healthy controls with a casecontrol study design.All cases were recruited from the department of Rheumatology and Immunology inpatient and outpatients in the Affiliated Hospitals to Anhui Medical University.The diagnosis criteria of SSc use the American College of Rheumatology in1980 criteria.The detection of the genotyping used Taq Man allele discrimination assay.Data analysis was performed by SPSS16.0,and P<0.05 was considered as statistically significant.Bonferroni correction was used for multiple comparisons.Results Genotype frequencies of IL-4-590C/T(rs2243250),IL-4R-Q576R(rs1801275),IL-13-1112C/T(rs1800925)in healthy were accorded with Hardy-Weinberg Equilibrium test.There were significant difference between SSc patients and healthy controls in allele frequencies of IL-4R-Q576R(rs1801275)(OR=2.286,95% CI: 1.212,4.313,?2=6.771,P=0.009).There were no difference in genotype and allele frequencies of IL-4-590C/T(rs2243250),IL-13-1112C/T(rs1800925),IL-13RA1-43163G/A (rs6646259)between SSc patients and healthy controls.Further analysis found that there were significant difference in IL-4-590C/T(rs2243250)polymorphism between SSc and controls in recessive model and homozygous model(?2=6.688,P=0.010;?2=6.475,P=0.011).For the relationship between gene polymorphism and SSc clinical features,we found that the allele frequencies of IL-4-590C/T(rs2243250)were associated with joint involvement in SSc(?2=10.694,P=0.001).Conclusions Our research showed the G allele of IL-4R-Q576R(rs1801275)was associated with SSc in Chinese Han population.
