Effect Of Huoxue Tongdu Decoction On The Neurological Repair After Sciatic Nerve Cutting Sutured In Rat

ObjectiveTo observe the effect of Huoxue Tongdu Decoction on function recovery after sciatic nerve cutting repaired in rat,and to explore its possible mechanism.Methodsthe right sciatic nerve of all the 40 rats were cut off and then were matched in situ,after that they were randomly divided into normal saline group(n=20)and Huoxue Tongdu Decoction group(n=20).For 4 weeks of post-operation,Huoxue Tongdu Decoction group was gavaged with Huoxue Tongdu Decoction twice a day,while normal saline group were both gavaged with normal saline twice a day.Weekly detecting sciatic nerve function index,taking the appropriate tissue after four weeks:Triceps muscle of calf were completely cut,then be weighed and recorded;each 10 rats of each group were separately taken the injured sciatic nerve for morphology and protein science test.Morphology test included HE staining to observe the injured sciatic nerve morphology,and immunohistochemistry to detect the distribution and expression of BDNF and NT-3 in the injured sciatic nerve;Protein BDNF and TrkB in the injured sciatic nerve were detect by Western blot technique for semi-quantitative analysis.ResultsDuring the 4 weeks after sciatic nerve cutting repaired,SFI in both Huoxue Tongdu Decoction group and normal saline group were gradually increased,suggesting function were restored in both 2 groups,but the Huoxue Tongdu Decoction group increased significantly compared with the normal saline group(P<0.01);At the time of 4 week,compared the wet weigh of calf triceps muscle of the injury side in the 2 groups,Huoxue Tongdu Decoction group is significantly heavier than normal saline group(P<0.01).On morphological manifestations,it can be seen that the density of nerve fiber in Huoxue Tongdu Decoction group is higher than normal saline group;Immunohistochemistry staining shows that BDNF and NT-3 distributed in the Schwann cells,and the average optical density of BDNF and NT-3 in Huoxue Tongdu Decoction group is significantly higher than normal saline group(P<0.01).The amount of BDNF and TrkB detected by western blot showed that BDNF in Huoxue Tongdu Decoction group is significantly higher than normal saline group(P<0.01),while TrkB in Huoxue Tongdu Decoction group is higher than normal saline group(P<0.05).ConclusionBased on sciatic nerve cutting repaired,the application of Huoxue Tongdu Decoction group has a role in promoting functional recovery.This is related to increasing expression of BDNF and NT-3 in Schwann cells of the nerve fiber,and play the role through the BDNF-TrkB way.
