Objective: 1.To evaluate the efficacy of the PowerPlex? Fusion kit as a forensic genetic marker in the Guizhou Shui population;2.To explore the genetic differences between Guizhou Shui and other 16 ethnic groups.Methods: 1.Blood samples were obtained from 400 apparently unrelated blood donors from the Shui population living in Guizhou provinceand blood samples were extracted by Chelex-100 method;2.DNA was directly amplified using PowerPlex? Fusion kit,and PowerPlex?Y23 kit.The amplified PCR products were separated using the 3500 Series Genetic Analyzer and genotyped according to GeneMapper ID-X software;3.Amplification,sequencing,splicing and polymorphism named the mtDNA;4.Analyze the genetic polymorphism data of each group and the genetic structure of the population by Powermarker,WinArl3.5 and MAGE4.0.Results: 1.The result showed that 238 alleles were observed in 300 samples.The allele frequency distribution was 0.0036~0.5324;2.The heterozygosity(H)was 0.5927~0.8907.The polymorphism information content(PIC)was 0.5314~0.8819.The discrimination power(DP)was 0.7605~0.9778.The power of exclusion(PE)was 0.2822~0.7765.The match probability(Pm)was 0.0222~0.2395.The combined match probability(CPm)was 3.4968×10-26;3.A total of 173 Y-STR haplotypes were observed in the 200 samples,of which 154 were unique,19 shared haplotypes.The chromosome haplotype difference(HD)was 0.9993.A total of 75 mtDNA haplotypes were observed from 80 samples,of which 70 were unique and 5 shared haplotypes.The haplotype difference(HD)of the mtDNA locus was 0.9984;4.From the autosomal’s datas there was no obvious difference between Guizhou Shui and Guangxi Han,Guangxi Zhuang,Guangxi Yao.The allele frequency distribution of Guizhou Shui and other ethnic groups was different(p<0.05).From the Y chromosome’s datas,the allele frequency distribution of Guizhou Shui and other ethnic groups was different(p<0.05);5.The autosomal’s phylogenetic tree consists of two main branches.One includes Guizhou Shui and other 14 ethnics,Yunnan Zhuang build up the other main branches.The Y chromosome’s phylogenetic tree consists of two main branches,too.One includes Guizhou Shui and other 9 ethnics,Guangxi Yao build up the other main branches.Conclusion: 1.The PowerPlex? Fusion kiti had high polymorphism in Guizhou Shui.They were powerful and reliable for forensic personal identification,paternity testing;2.The results of the study of ethnic origin of Guizhou Shui have further supported the ‘From Guangxi and Guangdong’;3.The genetic relationship between Guizhou Shui and other ethnic groups is basically consistent with its language and language,geography and historical data;4.The results will promote the study of ethnic origin of Guizhou aquarium,and enrich the research data of ethnic groups in our country,and provide information for the inheritance relationship among ethnic groups or ethnic groups.
