objective: 1.To analyze the gene chip of ischemic cardiomyopathy by bioinformatics tools to choose screening significant genes,for subsequent experimental study;2.To study the expression of hypoxia inducing factor 1 alpha to the pathogenesis of ischemic cardiomyopathy independent effect size in eliminate confounding factors and the effect after the modification factor and provide new ideas for the etiological studies of ischemic cardiomyopathy in forensic pathology identification.Method : 1.At the National Center for Biotechnology Information(NCBI)public microarray database(GEO)gene in ischemic cardiomyopathy chip data download,data processing standard edition,using Limma(R language package)significant P statistic calculation of ischemic cardiomyopathy group ICM and normal group NF values by Benjamini.2.The autopsy cases in 2013-2015 were collect from forensic judicial authentication center,guizhou medical university.The standard criteria were the pathology confirmed that there were varying degrees of coronary artery stenosis,or had already diagnosed as suffering from coronary heart disease.The exclusion criteria were merging other types of cardiomyopathy;and death and the autopsy time > 72 hours.3.With ischemic cardiomyopathy happen whether as a group,through independent sample t-test,mann? wheater U test and chi-square test,statistical methods,analysis of relevant clinical features is described.4.According to the diagnostic criteria,myocardial tissues were observed by microscopy and the expression of hypoxia inducible factor alpha-1 in myocardial cells was tested by immunohistochemical staining.5.The multiple regression equation model was established,the independent effect of hypoxia inducible factor-1 alpha and ischemic cardiomyopathy value were evaluated.Result:1.The differences were found in gene1358,in which were involved in cell cycle,gap link,P53 and other biologicalprocesses and signaling pathways.The significantly genes differentially expressed protein interaction network were likely built and HIF1 alpha gene was located in the center of the network;2.Confounding factors and effect modifiers under controlled conditions,the independent effect of HIF-1a on ischemic cardiomyopathy: the HIF-1a negative expression group as a reference,the probability of occurrence of ischemic cardiomyopathy disease positive samples did not increase(P >0.05).Compared with negative samples,the incidence of ICM increased 3.7 and 7.5 times(model 1)in the positive and strong positive samples.The results of model 2 showed that OR's value and direction were more robust,increased 4.2 and 8.4 times,respectively.Conclusion: 1.The application of bioinformatics software can analyze one of the key proteins screening of ischemic cardiomyopathy in the microarray data.2.HIF-1 a in ischemic cardiomyopathy in the development of the role of protein interaction plays a key link,it and other surrounding are close relations between gene product,further illustrate ischemic cardiomyopathy development is the result of a variety of protein interaction;3.The alpha HIF-1 associated with the occurrence of ICM.
