Objectives : To determine the expression of hypoxia-inducible factor-1α(HIF-1α) in children with wilms tumor (WT) ,and its correlation with various clinicopathology parameters, microvessel density(MVD) and prognosis.Methods : Select 45 cases of nephroblastoma resecting specimens in the First Affiliated Hospital of Guangxi Medical University Pediatric Surgery from January 2003 to January 2008, which is confirmed after surgical pathologically. Group them respectively by age, sex, histopathological type, clinical stage and NWTS risk factors. Using immunohistochemical streptavidin peroxidase conjugated method test the expression of HIF-1αand CD34 in WT organizations and adjacent tissues. Calculate MVD by CD34 staining. Then combined with the clinical prognosis of follow-up results of the experimental data and determine HIF-1α's correlation with various clinicopathology parameters, MVD and the cumulative survival rate.Results : In nephroblastoma tissues,HIF-1αexpressed in high levels, but low levels in adjacent tissues, and the intensity of expression of HIF-1αwas increased with the increasing of clinic stages of the tumor(P<0.05).The positive expressions of HIF-1αin different pathological types are no difference (P>0.05).The intensity of expression of microvessel density was increased with the increasing of clinic stages of the tumor, and the expression of micrevessel density was found to be more intense in advanced stages of tumor (P<0.05). Microvessel density in HIF-1αpositive group is larger than in negative group (P<0.05). The expression of HIF-1αin high risk group is more intense than low risk group (P<0.05). The live time of low risk group is longer than high risk group.Conclusion : (1) The expression of HIF-1αin WT was significantly higher than tumor tissue, suggesting that it may be related highly to the growth of WT; (2) The expression of HIF-1αin the WT was positively correlated with MVD. That may contribute to WT angiogenesis; (3) The expression of HIF-1αmay indicate a poor prognosis of WT, it may judge the value of the prognosis.
