Objective:To establish and evaluate a new animal model of menopausal syndrome which more close to human menopausal transition pathological changes by using the method of chronic unpredictable stimulation to natural aging female rats;Use Shu Shen Tiao Geng Fang treat rats with menopausal syndrome,to understand the effect of this prescription on the sex hormone level and the morphological effects of the reproductive system in rats with menopausal syndrome,to explore the mechanism of menopausal syndrome in the process of treatment and provide theoretical basis for treatment of menopausal syndrome in clinical work.Methods:Forty male SD rats aged 11-12 months were randomly divided into blank control group,lower dose Shu Shen Tiao Geng Fang group,higher dose Shu Shen Tiao Geng Fang group,western medicine control group 4,10 rats aged 4months as young control group,40 early age rats were subjected to chronic unpredictable stimulation for 2weeks.The specific methods included swimming,fasting,ban,bound water,shaking,clip the tail.After the end of the stimulus,test the success of the model,the test method: daily observation of rat vaginal exfoliative cell smear,for 2 weeks,to understand its oestrous cycle,cycle disorder,interphase extended indicated building success.The rats in the four groups were treated with normal saline,low dose Shu Shen Tiao Geng Fang,high dose Shu Shen Tiao Geng Fang and estradiol valerate solution.The young rats Given normal saline.After 20 days of drug treatment,gather the eyeball blood make 5 ml,broken neck death,rapid cutting in rats with bilateral ovaries,uterus,measure the serum estradiol?E2?Progesterone?P?,follicle-stimulating hormone?FSH?and luteinizing hormone?LH?.calculate the The ovaries and uterus indexes.Results:1.The use of chronic unpredictability to stimulate the role of the old rats in this method is relatively successful to establish a relatively consistent with the performance of human perimenopausal syndrome rat model is more reliable to establish perimenopausal syndrome rat model Method,worthy of promotion.2.Compared with the young group,lower dose Shu Shen Tiao Geng Fang group,higher dose Shu Shen Tiao Geng Fang group and western medicine control group had lower E2,P,uterine index and ovarian index,and the difference was significant?P<0.05?,while FSH and LH were significantly increased?P<0.01?.3.Compared with the blank control group,the levels of serum E2,P,uterus index and ovarian index in the lower dose Shu Shen Tiao Geng Fang group,higher dose Shu Shen Tiao Geng Fang group and western medicine control group were significantly lower than those in the blank control group?P<0.01?.The FSH and LH were decreased,difference was significant?P<0.05?.4.Compared with the high dose group,serum E2,P,uterus,ovarian index are decreased in low dose group and western medicine control group with significant difference?P<0.05?;FSH,LH are increased with obvious difference?P<0.05?;The western medicine control group of FSH,LH are increased with significant difference?P<0.01?.5.Low dose group compared with the western medicine control group,there's no difference between the two groups of serum E2?P>0.05?;P,uterus,ovarian index are obviously different?P<0.05?;And the level of FSH,LH have significant difference?P<0.01?.Conclusions:1.The chronic unpredictability stimulation in natural aging rats is successfully established rat model of menopausal syndrome which is relatively conforms to human performance,is a more reliable method which is worth to promoting.2.Shu Shen Tiao Geng Fang has good control effect in menopausal syndrome rats' sex hormone levels,uterus,ovarian index,provides a theoretical basis for clinical work in the treatment of menopausal syndrome,which worthy of clinical popularization and application.
