Chinese herbal formulas is a performance of syndrome identifying treatment variation and holism concept.As we all known,Chinese herbal formulas are constituted according to way of assistant and guide.Chinese herbal formulas includes many kinds of traditional Chinese medicines,each medicine has hundreds of chemical components and each component has different target,the trait above make it difficult to study the Chinese herbal formulas.From the pharmacokinetic perspective,it was considered that compounds absorbed into the blood have a chance for showing bioactivities,we should not only research the components in extraction of BaoErKang,but also study the components exposing in plasma.In order to illuminate the material base of pharmacodynamics and the law of metabolism in vivo,a High-resolution mass spectrometry method is used to identify the main components in BaoErKang.The components in BaoErKang are studied after oral administration of BaoErKang to dog.Several main components are chosen as marker components after study of components absorbed in blood,and a HPLC-MS/MS methods is used to quantify the maker components,the ideals of research are as follows: Chapter one: the summary of related literature of BaoErKangChapter one illuminates the drug matching and adaptation disease,it also introduces the place where the BaoErKang is mostly used.The main feature and main effects of BaoErKang are introduced in this chapter,in addition,a summary of components of each traditional Chinese medicine is conducted according the related literature.Chapter two: the research of components in BaoErKangTechnology of UPLC-QTOF is used to identify the main components in extraction of BaoErKang,seven components are identified by comparing with the relative literatures,including Naringin,Ononin,Liquiritin,Formononetin,3,5,6,7,8,3',4'-heptemthoxyflavone,Atractylenolide?,Dehydroeburicoic acid.Nine components are assured by comparing with the corresponding reference standards,including Hordenine,Scopoletin,Vitexin,Hesperetin,Liquiritigenin,Hesperidin,Glycyrrhizic acid,Atractylenolide ?,Glycyrrhetic acid.The analysis of components in BaoErKang and application of UPLC-QTOF technology provide the information for the study of components exposing in the blood.Chapter three: the research of components exposing in bloodOn the base of study of components in the BaoErKang,UPLC-QTOF technology was applied to research the components exposing in blood after oral administration of BaoErKang to dog,three components are identified by comparing with the relative literatures,including Naringin,Ononin,Atractylenolide?,Dehydroeburicoic acid.Nine components are assured by comparing with the corresponding reference standards,including Scopoletin,Vitexin,Hesperetin,Liquiritigenin,Hesperidin,,Atractylenolide?,Glycyrrhetic acid.Chapter four: quantification of biomarkers exposing in bloodAn effective method is created to quantify the maker components exposing in the blood and the maker components are chosen according the result of research of components in blood.Three components are quantified simultaneously,including Hesperidin,Liquiritigenin and Scopoletin.Method validation includes lineary,accuracy,precision,matrix effect and so on.The method is applied to quantify the concentration of biomarkers in defferent time,and is considered as a effective way to quantify the concentration of biomarkers.The UPLC-High-Resolution mass spectrometry technology is applied tostudy the material base of BaoErKang,HPLC-MS/MS method is applied to quantifiy the marker components in blood.The information of quality control is provided by researching the components in BaoErKang,The UPLC-High-Resolution mass spectrometry technology successfully analyzes the components in extraction of BaoErKang and plasma of dog,it has the trait of rapid analysis,simple process and good repetition,the study of BaoErKang provides the scientific infomation for the futher research.
