Objective:To study the effects of leeches containing plasma and serum on the proliferation,migration and VEGF of human pancreatic cancer PANC-1cells and to compare the mechanism of leeches containing plasma and serum in human pancreatic cancer PANC-1 cells.Research to provide experimental basisMethod: 16 SD rats were randomly divided into two groups.The experimental group was fed with 5ml/Kg of the concentration of0.2315 g / ml of leech Chinese medicine decoction twice a day for 4 days.The blank control group was fed with the same amount Saline,1% pentobarbital sodium anesthesia,abdominal aorta blood collection,were prepared with plasma,serum,blank plasma,serum,with CCK-8 method to detect plasma containing plasma,serum on cell proliferation inhibition,cell The expression of VEGF in VEGF was detected by immunofluorescence and Western Blot.The expression of VEGF was detected by Rt-PCR.Results: 1.The results of CCK-8 showed that the proliferation rate of human pancreatic cancer PANC-1 cells was slower than that of the control group(p<0.05).The inhibitory rate of the plasma group was significantly higher than that of the control group(p<0.05).The inhibitory rate of the plasma group was significantly higher than that of the control group(p<0.05).2.The results showed that the sclerosis healing rate of the blank control group was relatively slow compared with that of the drug-containing serum group and had a certain inhibitory effect on the migration of human pancreatic cancer PANC-1 cells(p<0.05).The inhibitory effect on the migration ability of PANC-1 cells was stronger than that of the drug-containing serum group(p<0.05).3.Immunofluorescence,RT-PCR and Western Blot showed that the expression of VEGF in human pancreatic cancer cell line PANC-1 was significantly lower than that in the control group(p<0.05).In the immunofluorescence assay and Western Blot experiment,the plasma containing plasma group had obvious effect on the expression of VEGF in the serum group.Conclusion: 1.The plasma and serum of the leeches had a strong inhibitory effect on the proliferation of human pancreatic cancer cell line PANC-1,and the inhibitory rate increased with the prolongation of time,and the inhibitory effect of the drug-containing plasma on the cells was stronger than that of the drug-containing serum.2.The migration of Pancreatic cancer cells and serum could inhibit the migration of human pancreatic cancer PANC-1,and the migration effect of pancreatic cancer cells was stronger than that of drug-containing serum.3.The expression of VEGF in human pancreatic cancer PANC-1 cells was down-regulated by plasma and serum,and the effect of plasma on VEGF was significantly higher than that of drug.4.The plasma inhibits pancreatic cancer cells with PANC-1 is stronger than that of drug-containing serum,suggesting that the plasma of leeches is more advantageous than the drug-containing serum.5.Inhibitory effect of leeches containing plasma and serum on the proliferation of human pancreatic cancer PANC-1 cells may be related to the down-regulation of VEGF expression...
