Objective:Through observing the serum levels of lipids,ALT,AST,FPG,FINS and MDA,SOD,T-GSH in liver of the rats,the liver tissues pathological changes,the expressi-on level of mRNA and protein of NF-?B,TNF-?,further discussed the molecular mech-anismabout the intervention of expelling damp,resolving phlegm and promoting blood circulation for nonalcoholic fatty liver disease in rats,providing theoretical basis and va-rious treatments for clinical treatment of nonalcoholic fatty liver disease.Method:The sixty Wistar rats were randomly divided into the normal group and model group.Except the normal group,the rats in other groups were administered by high-fat emulsio besides basic diet to induce NAFLD model for 5 weeks,and the model group were randomly divided into:the model group,choline group(polyene phosphatidyl choline intervention group),Traditional Chinese Medicine(TCM)low dose group(Jiawei Yinshao pulvis low dose intervention group),TCM middle dose group(Jiawei Yinshao pulvis middle dose intervention group)and TCM high dose group(Jiawei Yinshao pulvis high dose intervention group).At the Sixth week,the rats in the choline group and the Traditional Chinese Medicine groups were individually treated with the daily intragastric administration of polyene phosphatidyl choline and the Jiawei Yinshao pulvis.After 12 weeks sacrificed,rats of each group were weighed and the liver tissues were collected to evaluate the liver weight;detected the serum levels of TG,TC,HDL-C,LDL-C,FFA,ALT,AST and MDA,SOD,T-GSH in liver of the rats;observed the liver tissues pathological changes by HE staining and Oil Red O staining.Detected each rat liver tissue NF-?B and TNF-a expression situation by Immunohistochemical,detected the expression level of mRNA of NF-?B and TNF-a by RT-PCR.Result:1.Compared with the model group,the serum TC,TG,LDL-C,FFA,FPG,AST,ALT,FIN S,H-OMA-IR levels and the liver index in TCM groups were significantly decreased(P<0.05),the serum HDL-C and the liver levels of SOD,T-GSH in TCM groups were signi-ficantly increased(P<0.05).2.HE staining showed that there was obvious adipose degeneration in the hepatocyte of model group,a large number of lipid droplets were seen in the field of vision,accompanied by a significant cytoplasmic loose fishnet degeneration,existed infiltration of inflammatory cells in the portal area.The damage degree of the Hepatocyte structure of TCM high dose group,TCM medium dose group and the number of the lipid droplets produced were less than the model group and the choline group.Oil Red O staining showed that the liver nuclei of rats were stained with dark blue,a large number of diffuse red lipid droplets appeared in the cells and the lipid droplets of hepatic cell have been fused to form large lipid droplets in the model group.Compared with the normal group,the content of lipid droplets in the liver of rats with the TCM high dose group was the least.3.The RT-PCR results showed that the expression lever of mRNA of NF-?B and TNF-?in hepatic tissue of TCM group was lower than model group and choline group.The Immunohistochemical results showed that the positive expression of NF-?B,TNF-? in the model group was higher than drug group,the positive expression of NF-?B,TNF-? of the choline group was higher than the TCM high does group.Conclusion:The Modified Yinshao pulvis which was based on expelling damp,resolving phlegm and promoting blood circulation,could alleviate the progression of nonalcoholic fatty liver disease and had a better control effect on NAFLD.Its possible mechanism of action was:1.The Modified Yinshao pulvis which was based on expelling damp,resolving phlegm and promoting blood circulation,could improve lipid metabolic disturbance,reduce blood fat and the excessive deposition of lipid in the liver,decrease the degeneration and thanatosis of liver cells,and improve the role of liver function.2.The Modified Yinshao pulvis which was based on expelling damp,resolving phlegm and promoting blood circulation,could debase blood glucose and insulin levels to improve insulin resistance,thus reversing the severity of fatty liver.3.The Modified Yinshao pulvis which was based on expelling damp,resolving phlegm and promoting blood circulation,could improve the ability of the liver against oxidative stress and lipid peroxidation to alleviate the progression of nonalcoholic fatty liver disease.4.The molecular mechanism about the intervention of Modified Yinshao pulvis for NAFLD was related to inhibition or down-regulation of the expression of NF-?B and TNF-?that blocking the activation of IKK?/NF-?B signaling pathway.
