Part ?: Expression and significance of miR-34/449 in peripheral blood of children with asthmaObjective: To investigate the expression of miR-34/449 in peripheral blood of children with acute exacerbation of bronchial asthma and its relationship with the condition of children.Method:A total of 48 children with acute exacerbation of bronchial asthma admitted to our hospital from January 2015 to January 2017 were enrolled in the asthma group and 40 healthy children were included as controls in the same period.The levels of serum eosinophils,IgE,IL-5,TNF-?,IL-13 and IFN-?were detected by microRNA-34 and microRNA-449 levels.Results: The levels of microRNA-34 and microRNA-449 in the asthma group were(0.26± 0.12)and(0.38 ± 0.07)respectively.The levels of microRNA-34 and microRNA-449 in the control group were(0.98 ± 0.19),(1.03 ± 0.21),The difference was significant(P <0.001).The levels of eosinophils,IgE,IL-5,IL-13,TNF-? and IFN-? in asthmatic children were(0.45 ± 0.12)109 / L,(169.47±100.58)IU/mL,(16.85±3.31)pg/m L,(57.56±20.89)pg/m L,(15.57±3.07)pg/mL,(65.69±10.31)pg/mL,the number of eosinophils in children,IgE,IL-5,IL-13,TNF-? and IFN-? levels in the control group were(0.23± 0.09)× 109 / L,(72.47 ± 32.58)IU / mL,(8.75 ± 1.57)pg / mL,(8.80 ± 9.51)pg / mL,(8.12 ± 1.50)pg / m L,(77.38 ± 9.23)pg / m L,the difference was significant(P <0.05).Conclusion: miR-34/449 is closely related to the development of asthma.It is worthwhile to improve the expression of Mir the study.Part ?: Effect of expression of miR-34/449 on airway inflammation and airway remodeling in asthmatic mice and its correlation with autophagyObjective: To study the effect of expression of miR-34/449 on airway inflammation and airway remodeling in asthmatic mice and its correlation with autophagy,and to lay the foundation for the application of miR-34/449 in the treatment of asthma.Methods: Eighteen male BALB/c mice aged 6-8 weeks were randomly divided into asthma model group,normal control group,miR34/449 mimics treatment group,6 mice in each group,The model of asthma was induced by intraperitoneal injection of sensitized and aerosolized inhaled mice,and The levels of IL-5,IL-6 and TNF-? in bronchoalveolar lavage fluid(BALF)were detected by ELISA.The lungs were observed by HE staining.The expression of miR34/449 in lung tissue was detected by RT-PCR.The expression of TGF-? was detected by immunofluorescence.The levels of bFGF,collagen I,collagen III and ?-SMA were detected by Western blot.The expression of LC3-II/LC3-I,Beclin-1 and P62 was observed,and the mechanism of the expression of miR-34/449 on airway inflammation and airway remodeling in asthmatic mice and its mechanism was discussed.Results: The infiltration of inflammatory cells in lung tissue of asthmatic mice model group was mainly in eosinophils,the levels of IL-5,IL-6 and TNF-? in BALF,TGF-?,bFGF,collagen ?,The level of ?-SMA increased,while the expression of autophagy associated protein LC3-II/LC3-I,Beclin-1 increased.The expression of mi R34/449 in lung tissue of asthmatic mice model group was lower than that in normal group.The expression level of miR34 / 449 in mi R34/449 mimics group was significantly higher than that in normal group and asthmatic model group(P <0.05).The levels of IL-5,IL-6 and TNF-? in BALF,TGF-?,b FGF,collagen I,collagen in lung tissue of mice treated with miR34/449 mimics group compared with asthma model group(P <0.05),and the expression of LC3-II / LC3-I and Beclin-1 decreased significantly(P <0.05).Conclusion: The expression of Mir-34/449 in the lung tissue of asthmatic mice decreased,and the overexpression of Mir-34/449 could significantly reduce the airway inflammation and reduce airway remodeling in asthmatic mice.This effect may be related to the decrease of autophagy Related protein expression.
