To explore the therapeutic effects and PET-CT in vivo tracing 18F-FDG labeled rat islets and bone marrow mesenchymal MSCs(MSCs)co-transplantation on diabetes rat models.Rat islets and MSCs were isolated and co-cultured in vitro.Third-passage(P3)MSCs for transplantation.Intraperitoneal injection of streptozotocin(STZ)for T1 DM model.The diabetes rats were randomly divided into four groups(n=5)for transplantation: group A(5000IEQ/kg islets+3*10^6/kg MSCs),group B(5000IEQ/kg islets),group C(3*10^6/kg MSCs)and group D(control).The C-peptide and insulin secretions and blood glucose in vitro or in vivo were determined.PET-CT was used to trace in vivo.Islets and MSCs treatment significantly enhanced islets function at 1 month after transplantation,as compared with islets without MSCs.The morphology of cells,C-peptide,insulin secretions and blood glucose of islets and MSCs in vitro or in vivo were all significantly better than islets treatment.Furthermore,PET-CT tracing in vivo have confirmed this.The effect of co-transplantation in the treatment of diabetes mellitus was better than that of single islet transplantation,and the PET-CT can reveal the distribution of islet and BMSCs in liver via portal vein transplantation.
