During the last century,rapid social development and changes in human lifestyle have resulted in a dramatic increase in the prevalence of diabetes mellitus in the world,which is a serious threat to human health.Though oral hypoglycemic agents and insulin are the main ways of treatment for diabetes,they have somewhat side effects.So it is very urgent for us to develop anti-diabetic plants for its less toxicity and no-drug-resistance.Perilla has more than 2000 years of cultivation history in China,mainly produced in Guizhou,Jiangsu,Anhui,Hunan and other places,with a very wide range of resources.In recent years,Perilla has attracted much attention of the whole world because of its unique active substances and nutrients,which has high economic value.Studies have shown that Perilla leaves extract can play an important role in the treatment for diabetic syndrome,but it still has no clear conclusion currently about the mechanism of these active ingredients.In this study,the hypoglyecemic effect of Perilla leaves extract and its related mechanism have systematically studied.Firstly,the research is designed to study the optimum extraction conditions of Perilla leaves,which were studied through the single factor experiment and response surface experiments.The results show that when the temperature is 52℃,extraction time is 60 mins,the extraction ratio is 1:25,and the extraction times is 3,AGI active ingredient in Perilla leaves have the highest rate.Secondly,the inhibition of Perilla leaves and acarbose to different sources of a-glucosidase were compared,the results showed that Perilla leaves have strong inhibitory activity on a-glucosidas which comes from porcine pancreatic intestinal,rice and yeast,while showes no inhibitory activity on a-glucosidase that comes from bacteria.On this basis,the inhibitory kinetic of Perilla leaves to different sources of a-glucosidase were measured.The results showed that the inhibition type of Perilla leaves on a-glucosidas which comes from porcine pancreatic intestinal,rice and yeast was competitive inhibition.Moreover,the inhibitory potency of samples on glucose transport was assessed in Caco-2 cell monolayer system.Whether Perilla leaves can inhibit the activity of enzymes that are related to digestion of carbohydrates was studied;then whether Perilla leaves can inhibit glucose transport under different blood sugar condition was also studied.The results show that Perilla leaves can inhibit both maltase and sucrase,and also has influence on glucose transporting. |
