Objective:To investigate the influence of blood glucose(BG)and glycosylated hemoglobin(HbAlc)levels on cerebral infarction.Methods:88 cases of cerebral infarction patients,according to the fasting blood glucose and previous have no history of diabetes mellitus were divided into diabetes mellitus(DM group,58 cases)group and non diabetes group(NDM group,30 cases),and select the hospital check-up 40 healthy control group(Control group,cases).Analysis of the correlation between blood glucose and glycosylated hemoglobin in patients with acute ischemic cerebrovascular disease.We developed high-fat diet-fed streptozotocin(HFD-STZ)rat,a well-known type 2 diabetic model,to evaluate the e□ect of BG and HbA2c on cerebral infarction.The mice were randomly divided into five groups(n = 12/group):Control rat group,NDM rat group,DM rat group,Pioglitazone(a PPARy Agonist)group and Combined(Pioglitazone+GW9662,GW9662 is a PPARy inhibitor)group.TTC staining method were used to observe the degree of necrosis in the brain of SD rats.Blood glucose and glycosylated hemoglobin levels were measured by blood glucose meter and ELISA method,respectively.The levels of interleukin-6(IL-6)and tumor necrosis factor-a(TNF-a)were analyzed to determine the inflammation status.The protein expression degrees of peroxisome proliferator-activated receptor gamma(PPARy)nuclear factor-k-gene binding(NF-κB p65)were assayed by Western blott.Results:Compare with control group and NDM group,the BG and HbA1c of DM group were significantly increased[(11.653.41)mmol/L vs(5.690.59)mmol/L,(4.230.52)mmol/L;(9.132.21)%vs(5.570.27)%,(4.030.53)%,P<0.05].Compare with control group and NDM group,The degree of cerebral infarction in DM group were significantly increased.Animal experimental results also showed that the cerebral infarct size in NDM rat group and DM rat group rats were higher than that in control rat group,and the cerebral infarct size in DM rat group was higher than NDM rat group.The pretreatment with pioglitazone decreased the cerebral infarct size.Compare to NDM rat group,the NF-κB p65 expression levels of DM rat group was upregulated,the PPARy expression levels of DM rat group was downregulated.Compare to NDM rat and DM rat group,the NF-κB p65 expression levels of Pioglitazone group was downregulated,the PPARy expression levels of Pioglitazone group was upregulated.And these effects of pioglitazone was reversed by co-treatment with GW9662.Conclusion:BG and HbAlc are closely related to cerebral infarction,and hyperglycemia may aggravate cerebral infarction injury.
