| Objective: Establish rat endometriosis model by autologous transplan--tation method(EMs)model,and then divided into 6 groups: model group,Guizhi Fuling Capsule low,middle and high dose group,Sanjie capsule group,mifepristone group were given normal saline,Guizhi Fuling cap--sule low,and high dose suspension,Sanjie capsule suspension and mifepristone suspension,To observe effect of Fang Guizhi Fuling Pill which was called the behalf of blood and water on rat ectopic endometrial morphology,pathology and intervention effect on the expression of apoptosis factor caspase-3 and survivin with establishing rats endometriosis model by autologous transplantation method.To explore the possible mechanism of Guizhi Fuling pill in the treatment of EMs and explain academic connotation of blocked blood formed water which led EMs.Methods: 1.Establishing rats endometriosis model: female SD rats were selected in 150 cases,vaginal pictures,HE staining,the establishment of estrous cycle files.The experiment was carried out in 135 SD rats with regular estrous cycle,15 rats in the blank group,15 in the sham operated group(n=9),and the remaining 105 rats were implanted with EMs method in the estrus.We prevent infection and promot growth of ectopic endome--trium by intramuscular injection of gentamicin and estradiolbenzoate.After 4 weeks,the rat model was observed by open surgery.2.Grouping:the rats which have been established model successfully were selected and randomly divided into model group,high,middle and low dose group of Guizhi Fuling Capsule,Sanjie analgesic capsule group and mifepristone group.3.Index test: Treatment with medicine for 4 weeks,We can observe the mor--phology and pathological changes of ectopic endometrium and the expression of apoptosis factor by open surgery.Results: 1.90 EMs models were successfully established by autologous transplantation method in this experiment.The rate of model was 85.71%.2.Histomorphology observation:In the model group,there were obvious hyperplasia of ectopic endometrium,the number of glands,the enlargement of glandular cavity,and the glandular epithelium was arranged in layers.In each medication group,ectopic endometrial glands were atrophic,the number of glands cells was decreased,cells were flat,and the arrangement of cells was loose and non polar.,especially in high group of Guizhi Fuling pill and mifepristone group.3.The expression of apoptosis factor in endometrial tissue: the expression of caspase-3 was low in ectopic endometrium of model group.The treatment group can increase the expression of caspase-3 in different degree.The difference was significant(P<0.05),especially in high group of Guizhi Fuling pill and mifepristone group.The expression of survivin in ectopic endometrium of model group was high.The treatment group can decrease the expression of survivin in different degrees,the difference was significant(P<0.05),the expression of survivin was the lowest in the high group of guizhi Fuling pill andmifepristone group.Conclusions:1.The EMs model was successfully established by using the method of autologous transplantation,and the rate of the model was85.71%.The morphology of ectopic endometrium accords with the standard of EMs model 2.The separation surface of endometrium is complete,the speed of separation is fast,endometrium is not easy teared or damaged by using the method of "water injection" for the separation of endometrium that can be used as a method to separate the endometrium of rat uterus.3.Guizhi Fuling pill can reduce the volume of ectopic lesions,inhibit the growth of ectopic endometrium,accelerate its atrophy,and necr--osis.It can treat EMs.The best treatment of EMs is high group of Guizhi Fuling pill.4.The mechanism of Guizhi Fuling pill in the treatment of EMs which is the representative prescription of Huoxue Lishui may be the high expression of survivin and the activation of cells in caspase-3activation pathway,which induces ectopic endometrial cells atrophy,necrosis and apoptosis,and plays a role in the treatment of EMs.So the blood stasis is the key mechanism of EMs. |
