| Aim:By comparing the effects of Danggui-shaoyao-san and its decomposed recipes on blood stasis rats,spleen deficiency rats with internal dampness arrest and primary dysmenorrheal mice models.This paper explores the differences of pharmacodynamic effects of Danggui-shaoyao-san and its decomposed recipes on different models,and preliminarily explores the connotation and characteristics of Danggui-shaoyao-san in promoting blood and water circulation.Methods:Firstly,this paper would,with the help of rat model,examine the acute blood stasis induced by adrenaline and ice-water bath,detection of hemorheological parameters:whole blood viscosity,plasma viscosity,erythrocyte sedimentation,hematocrit;the coagulation function indicators:prothrombin time(PT),activated partial thromboplastin time(APTT),fibrinogen(FIB)content;Lung wet/dry(W/D)ratio and AQP1 expression in lung/kidney tissues.Secondly,this paper would look into the reproduction of spleen deficiency rat model of water-dampness intermediate stopping by diet,overwork and water load.Detection of fecal water content,urinary D-xylose excretion,water load index,serum albumin,total protein and AQP1 expression in kidney tissue.Thirdly,this paper would checkthe estradiol valerate combined with oxytocin used to duplicate the primary dysmenorrheal,and analyze torsion number as well as the levels of Ca2+,PGE2,PGF2α,PGF2α/PGE2 in uterine tissues and the expression of AQP1.Results:(1)The hemorheological indexes of rats with acute blood stasis became higher,PT and APTT index decreased,FIB content increased.Each administration group can reduce the indexes of hemorheology and significantly improve the indexes of coagulation function.The total blood-activating effect of Danggui Shaoyaosan group was the best.The total blood-activating effect of the blood-activating drug group was similar to that of the Danggui-chuangxiong group.The blood-activating effect of the water-promoting drug group was reflected in APTT and hematocrit.The lung wet/dry(W/D)ratio of model rats was decreased in each administration group(P<0.05 or P<0.01).The expression of AQP1 in lung tissue of model rats was increased in Danggui-Shaoyao-San group(P<0.05),while the expression of AQP1 in kidney tissue was less in Danggui-Chuanxiong group(P<0.01).(2)In the model of rats with spleen deficiency and dampness retention,the anal temperature of rats did not change much,but the water content of feces increased.D-xylose excretion,6h urine volume,serum total protein and albumin decreased,and AQP1 expression increased in kidney tissue.After administration,D-xylose excretion,6h urine volume,serum total protein and albumin were increased,and AQP1 expression in renal tissue was decreased(P<0.05 or P<0.01).The indexes of improving spleen deficiency and water-dampness retention in the water-promoting group were slightly better than those in the blood-activating group,but there was no significant difference.(3)In the model of primary dysmenorrhea mice,each administration group could significantly reduce the number of writhing induced by oxytocin,and the contents of Ca2+,PGE2,PGF2αand the ratio of PGF2α/PGE2 in uterine homogenate.The expression of AQP1 in uterus tissue of primary dysmenorrhea mice increased,and the expression of AQP1 in uterus tissue of each administration group decreased in varying degrees(P<0.05 or P<0.01).The effect of Danggui-Shaoyao-San on the index of primary dysmenorrhea was better than that of other groups.The effect of blood-activating drug group and Danggui-chuanxiong group on alleviating primary dysmenorrhea was similar.Conclusions:The effect of blood-activating drugs group was the same as that of Danggui-Chuanxiong group.The blood-activating drug group has a certain effect on water diversion,and the water-promoting drug group has a certain effect on promoting blood circulation.It embodies the essence that promoting blood circulation is beneficial to diuresis and vice versa. |
