Anthocyanins is the main medicinal ingredients in black rice and it has many kinds of activities such as blood lipid regulation,anti-oxidation and anti-inflammatory which is widely used in food,pharmaceutical products and other fields.This study was designed to evaluate the efficacy of black rice to regulate cholesterol metabolism and oxidative stress in C57BL/6J mice fed high fat diet.Blood lipid,antioxidant related indicators and gene expression of cholesterol metabolism and oxidative stress were measured to detect the effect of black rice dietary intervention in mice fed high fat diet.The main content and results shown as follow:We analyzed the effect of cholesterol metabolism regulation in mice fed high fat diet intervened by black rice.Serum lipid index,cholesterol content in liver and gene expression of cholesterol metabolism were measured.The results showed that compared with controls the total cholesterol(TC)content and triglyceride(TG)in serum,cholesterol content in liver were significantly decreased,the high density lipoprotein cholesterol(HDL-C)were significantly increased.RT-PCR results showed that compared with controls,the expression of LXR-??SREBP-2?LDL-R?CYP7A1 mRNA were significantly increased and the expression of HMG-CoA-R mRNA were decreased in liver.The expression of ABCG5?ABCG8 ? ABCA1 mRNA were significantly increased and the expression of HMG-CoA-R and NPC1L1 mRNA were decreased in intestine.The results indicated that black rice keep the balance of cholesterol metabolism by inhibiting the endogenous cholesterol synthesis and promoting cholesterol efflux.We studied the effect of oxidative stress regulation in mice fed high fat diet intervened by black rice.The activity of antioxidant enzymes and malondialdehyde content in serum,liver and brain,gene expression of oxidative stress regulation were measured.The results showed that compared with controls the activity of superoxide dismutase(SOD),glutathione peroxidase(GSH-PX)and catalase(CAT)were significantly increased and the content of malondialdehyde were significantly increased in serum,liver and brain.RT-PCR results showed that compared with controls,the expression of Nrf2,SOD1,SOD2,CAT and GPX1 mRNA were significantly increased in liver.The results demonstated that black rice protect the body from oxidative stress injury by increasing the expression of Nrf2 gene and its downstream genes related antioxidant.
