Efficacy And Safety Of Exenatide Treatment In Type 2 Diabetes Mellitus

Objective:To evaluate the efficacy and safety of exenatide therapy in patients with type 2 diabetes mellitus(T2DM).Methods:52 cases of patients with T2DM were divided into two groups according to their treatment history:treatment naive group(patients without history of any drug treatment)and previous treatment group(patients treated by metformin and/or sulfonylureas for more than three months but failed to control blood glucose or had gradually increased body weight)Patients were treated with exenatide,placebo and exenatide,insulin detemir for 12 weeks respectively and fasting plasma glucose(FPG),2-hour postprandial blood glucose(2hPG),glycosylated hemoglobin(HbAlc)and body mass index(BMI)were measured and compared after treatment,and any incidence of adverse reactions was observed.Results:Compared with pre-treatment indexes,of treatment naive group,2hPG,waist circumference(WC)and TG were obviously decreased after 12-weeks placebo treatment(P<0.05);FPG,2hPG,HbA1c,HOMA-IR,BMI,WC,TC and TG were all obviously decreased while HOMA-IS was significantly elevated after 12-weeks exenatide treatment(P<0.05).For previous treatment group:FPG,2hPG,and HbAlc,etc.were significantly decreased after 12-week insulin detemir treatment(P<0.05),but there were no obvious changes in indexes such as BMI,WC,and blood lipids;FPG,2hPG,HbAlc,HOMA-IR,BMI,and TC were all obviously decreased while HOMA-IS was significantly elevated after 12-week exenatide treatment(P<0.05).No severe hypoglycemia event occurred.Conclusion:For patients of type 2 diabetes mellitus,with or without previoustreatment,exenatide treatment can effectively reduce their blood glucose and body weight,and the adverse reactions are rare.
