OBJECTIVE:To investigate the expression and significance of IL-17A-producing T cells(Th17 cells Tc17 cells and ??T17 cells)and IL-1?,IL-17A,and IL-6 cytokines in the peripheral blood mononuclear cells(PBMCs)of different condition of children with hand-foot-mouth disease.METHODS:Collection of 60 cases of children with HFMD,according to clinical disease,severe group of 20 cases,mild group of 40 patients;and select healthy people the same age 40 cases in the control group.Extract of peripheral blood mononuclear cells,IL-17A+CD3+CD4+?IL-17A+CD3+CD8+ and IL-17A+??TCR+ marks of Th17 cells,Tc17 cells and y 8 T17 cells detected by flow cytometry.Liquid chip analysis platform for detecting foot-and-mouth disease and control group of IL-1?,IL-17A,and IL-6 levels of cytokines.The mRNA expression of IL-1?,IL-17A and IL-6 were analyzed by quantitative real-time PCR(qRT-PCR),then comparison of these parameters in patients with that after treatment.RESULTS:Children with severe hand-foot-mouth disease in peripheral blood mononuclear cells of Th17 cells and content of y 8 T17 cells higher than the light group and the control group,while lower Tc17 cells;In peripheral blood in children with severe hand-foot-mouth disease cytokines IL-1?,IL-17A,and IL-6 and its mRNA content was relatively mild and normal control higher;In mild cases of peripheral blood cytokines IL-1?,IL-17A,and IL-6 and its mRNA in no significant difference compared with healthy control group(P>0.05),no statistically significant differences.In patients with severe phenomena of whose contents were significantly decreased after treatment;The levels of IL-1?,IL-17A and IL-6 in peripheral blood were positively correlated with IL-1?,IL-17A and IL-6 mRNA levels,the serum levels of IL-1?,IL-17A and IL-6 in peripheral blood were positively correlated with Th17 cells and ??T17 cells in peripheral blood mononuclear cells,and negatively correlated with Tc17 cells.CONCLUSION:Foot-and-mouth disease virus infection in children with his condition after the occurrence and development of IL-1?,IL-17A and IL-6,increased cytokine expression tips after virus attacks the body,cause intense inflammatory and anti-inflammatory responses.Possible participation of IL-17A+T cells and related cytokines and progression of patients with severe hand-foot-mouth disease.
