Objective: Hypertrophic scar is very common sequel of human dermal injures especially after burn-wound healing. Though the pathogenesis of hypertrophic scar is still unclear, more and more research show that the hyperplasy and pruritus of human scar have close correlation with substance P,but the effects and mechanism(s) of Substance P in human hyperplastic scar is not clear. The experiment will investigate it as below :1. To investigate the effect and dose-effect relationship of substance P to fibroblast and mast cell in human hyperplastic scar. 2. To investigate the effect of substance P to the proliferation gene and apoptosis gene of fibroblast.3. To investigate the tissue contents and intracellular location ofβ-catenin protein of fibroblast in hyperplastic scar,then to presume the role ofβ-catenin to hyperplasy of human hyperplastic scar .4. To investigate whether substance P can regulate theβ-catenin express or not ,then to approach the mechanism that how substance P promotes the proliferation of fibroblast in human hyperplastic scar.5. To make use of substance P receptor antagons to block the effect of substance P, then to approach the blockage effect and make the basement for new drug exploitation to treat hyperplastic scar.Materials and methods: Hyperplastic scar and normal skin tissue have been collected and fibroblast has been cultured. Then used then to do experiments.1. The expression of substance P nerve fiber in hyperplastic scar and normal skin were determined by immunohistochemistry, and the expression of substance P in cultured fibroblast from hyperplastic scar and normal skin were determined by immunohistochemistry. Cell counting and MTT method were used to study the effects of SP on the proliferation and activity of cultured fibroblasts. Labeled with 3H-proline was used to study the effects of substance P on the collagen metabolism of cultured fibroblasts. Light and electron-microscopes were used to study the morphology changes of cultured fibroblasts.2. Our study used various concentrations of substance P or various concentrations of calcium under various affect time to assay histamine release of mast cell using immunofluorescence.
