Objective:Taking functional dyspepsia mice as research objects, observe the impact of electroacupuncture at Zusanli on gastric emptying rate,SCF/c-kit signal pathway, gastrointestinal cells of Cajal and smooth muscle cell of FD mice,discuss the relationship between gastrointestinal motility of electroacupuncture at Zusanli (ST36) and the impairment of SCF/c-kit signal pathway and change of ICC and SMC, so as to partially clarify the efficacy machanism of electroacupuncture on functional dyspepsia.Methods:36 C57BL mice of female and male SPF level were randomly divided into 3 groups:blank control group,model group and electroacupuncture group,12 mice in each group. Except blank control group,the other groups were modeled,according to the hunger arrhythmia and L-arginine integrated modeling method. After modeling, Stopped food intake,but kept drinking water for 24 hours,and then randomly selected six mice from each group to feed each one 0.5g with semisolid paste,after 15 minutes killed by the way of taking off the neck,taked out stomach from abdomen to measure gastric residual rate to make sure success of modeling,residual mice of each group were stoped modeling and recorded weight,food intake and other symptoms,and then restored normal intaking,at the same time electroacupuncture group was given acupuncture therapy for 7 days.After treatment,each group was stopped food intake,but kept drinking water for 24 hours, and then feeded each one 0.5g with semisolid paste,after 15 minutes killed,and taked out stomach from abdomen to measure gastric residual rate,collected stomach tissue to make slice,used HE to observe pathological changes of FD,and then taked residual gastric antrum tissue to detect protein content of SCF and c-kit by Western blot and mRNA expression of SCF and c-kit by QPCR,and observe ICC and SMC by immunofluorescence.Results:1.Behavior indexrAfter modeling,compared with blank group,weight and food intake of model and electroacupuncture group mice were decreased(p<0.05).After treatment,compared with model group, weight and food intake of eletroacupuncture group mice were increased(p< 0.05).2.Gastric emptying rate:Compared with blank group,gastric residual rate of model and electroacupuncture group mice was raised(p<0.05).Compared with model group,gastric residual rate of electroacupuncture group mice was lowered(p<0.05).3.mRNA expression and protein content of SCF and c-kit:Compared with blank group,mRNA expression and protein content of SCF and c-kit of model group mice were decreased(p<0.05).Compared with model group,mRNA expression and protein content of SCF and c-kit of electroacupuncture group mice were increased(p<0.05).4.Immunofluorescence:Structure of ICC and SMC of blank group mice was normal.Optical density value and number of ICC and SMC of model group mice were decreased (p<0.05) and the structure was impaired.Optical density value and number of ICC and SMC of electroacupuncture group mice were increased(p<0.05) and the structure was normal(p<0.05).Conclusions:1.After treatment,compared with model group,the study of weight,food intake and gastric emptying rate of electroacupuncture group mice implies electroacupunture can improve gastrointestinal motility.2.Electroacupuncture can increase mRNA expression and protein content of SCF and c-kit,improve structure of ICC and SMC,and then increase gastrointestinal motility.
