OBGECTIVE:To observe the effect of Pingganfang on behavioral sensitizations.conditioned place preference (CPP) induced by ethanol in mice and the concentration of dopamine (DA), pancreatic (Glu), gamma-aminobutyric acid (GABA) in brain of mice,to discuss the intervention mechanism of Pingganfang on alcohol Addiction.METHODS:(1)By observing the resident time in the white box and black box of mice to evaluate the effect of Pingganfang on the developing of conditioned place preference (CPP) in mice induced by ethanol. (2)By recording the locomotor activity in mice to evaluate the effect of Pingganfang on the developing and expressing of behavioral sensitization in mice induced by ethanol.(3) By measuring the concentration of dopamine (DA), pancreatic (Glu), gamma-aminobutyric acid (GABA) in the mice mesolimbic dopamine system by ELISA to apperceive the effect of Pingganfang on the concentration of dopamine (DA), pancreatic (Glu), gamma-aminobutyric acid (GABA) in the mice brain.Results:1 Effect of Pingganfang on the conditioned place preference (CPP)to ethanol in mice(1)The results of natural mice preferred side test show that, in the 15min testing time, the residence time of mice in the black box is longer than the white box, The difference between the two was statistically significant(p<0.01).(2) The results of the test period show that, the residence time of mice in the ethanol group was longer than the saline group and Pre-Workout, The difference between the two was statistically significant(p<0.01).(3) The results of the test period show that, the residence time of mice in the Pingganfang group was no difference to the saline group and Pre-Workout.(4) The results of the test period show that, the residence time of mice in the Pingganfang+ethanol group was shorter than the ethanol group, The difference between the two was statistically significant(p<0.01),and there is no difference between the residence time of mice in the Pingganfang+ethanol group and Pre-Workout.2 Effect of Pingganfang on the behavioral sensitization to ethanol in mice(1) The locomotor activity times baseline in mice had no difference.(2) The test results of the developing period show that, there was no difference during the five testes between the Pingganfang group and the saline group about the autonomic activity times, the autonomic activity times of mice in ethanol group was more than saline group, The difference was statistically significant(p<0.01).(3) The locomotor activity times of mice in ethanol group who accepted ethanol challenge was more than saline group who also accepted ethanol challenge, The difference was statistically significant(p<0.01).(4) There was no difference between Pingganfang group and saline group those who accepted saline, ethanol, Pingganfang and Pingganfang+ethanol challenge about the locomotor activity times of mice.(5) The locomotor activity times of mice in Pingganfang+ethanol group who accepted ethanol challenge was less than ethanol group, The difference was statistically significant(p<0.01).(6) The locomotor activity times of mice in ethanol group who accepted Pingganfang+ethanol challenge was less than who accepted ethanol challenge, The difference was statistically significant(p<0.01).3 Effect of PG on DA, Glu, and GABA levels in the brain tissues of mice of behavioral sensitizations to ethanol(1) The concentration of DA and Glu in the mice brain of ethanol group who accepted ethanol challenge was obviously more than saline group who accepted ethanol challenge,and the concentration of GABA was obviously less than it, the difference was statistically significant(p<0.01).(2) The concentration of DA, Glu and GABA in the mice brain was on difference between the Pingganfang group who accepeted Pingganfang challenge and the saline group who accepeted saline challenge.(3) The concentration of DA and Glu in the mice brain of Pingganfang+ethanol group who accepted ethanol challenge was obviously less than ethanol group who accepted ethanol challenge,and the concentration of GABA was obviously more than it, the difference was statistically significant(p<0.01).(4) The concentration of DA and Glu in the mice brain of ethanol group who accepted Pingganfang+ethanol challenge was obviously less than ethanol group who accepted ethanol challenge.and the concentration of GABA was obviously more than it, the difference was statistically significant(p<0.01).Conclusion:1 Effect of Pingganfang on the conditioned place preference (CPP)to ethanol in mice(1) Mice has a natural preference to the box with black floor and rough subface box, therefore, we adopt a biased experimental design and select the white box as drug-paried side, the black box as none drug-paried side.(2) Repeatedly treated (ig) with ethanol (2.2g/kg) can establish stable conditioned place preference model to mice.(3) Pingganfang does not influence the choice of mice about the natural preference side, reminding that Pingganfang has no rewarding effect.(4) Pingganfang can reduce the residence time of mice repeatedly treated (ig) with ethanol (2.2g/kg), indicating that Pingganfang can inhibit the developing of conditioned place preference to mice. That is to say, Pingganfang can inhibit the positive reward of ethanol.2 Effect of Pingganfang on the behavioral sensitization to ethanol in mice(1) The locomotor activity times baseline in mice had no difference.(2) Repeatedly treated (ig) with ethanol (2.2g/kg) increase the locomotor activity of mice. Repeatedly treated (ig) with Pingganfang has no influence about the locomotor activity of mice, indicating that Pingganfang has no addiction and sedation. Pingganfang was given 30min before ethanol(ig), and it can reduce the increase of locomotor activity times induced by ethanol, indicating that Pingganfang can inhibit the developing of the behavioral sensitization induced by ethanol in mice.(3) Repeatedly treated (ig) with ethanol (2.2g/kg) can establish stable behavioral sensitization model induced by ethanol to mice.(4) Repeatedly treated (ig) with Pingganfang can not induce behavioral sensitization induced to mice, That is to say, Pingganfang has no addiction.(5) Pingganfang given (ig)30min before ethanol (2.2g/kg) can inhibit the developing of the behavioral sensitization induced by ethanol to mice.(6) In the challenge phase,Pingganfang given (ig)30min before ethanol (2.2g/kg) can inhibit the expressing of the behavioral sensitization induced by ethanol to mice.3 Effect of Pingganfang on DA, Glu, and GABA levels in the brain tissues of mice of behavioral sensitizations to ethanol(1) Repeatedly treated (ig) with ethanol (2.2g/kg) can increase the concentration of DA and Glu.and decrease the concentration of GABA in the brain of mice.(2) Pingganfang has no effect on the concentration of DA, Glu and GABA in the brain of normal mice.(3) Pingganfang can inhibit the increase of DA and Glu,and the decrease of GABA to intervene in the development of behavioral sensitization to ethanol in mice.(4) Pingganfang can inhibit the increase of DA and Glu,and the decrease of GABA to intervene in the expression of behavioral sensitization to ethanol in mice.
